Small ubiquitin-related modifier (SUMO) fusion system has been shown to be efficient for enhancing expression and preventing degradation of the target protein. We showed herein that SUMO fusion to human keratinocyte growth factor 2 (hKGF-2) gene was feasible and it significantly enhanced protein expression and its efficiency. The fusion DNA fragment composed of SUMO gene, which was fused to hexahistidine tag, and hKGF-2 gene was amplified by PCR and inserted into the expression vector pET28a to construct the recombinant plasmid, pET28a-SUMO-hKGF-2.

The mutant of human basic fibroblast growth factor (hbFGF), hbFGF(Ser25,87,92), which was constructed by replacing the cysteine residues at the positions of the 25th, the 87th and the 92nd with serine residues, was coupled to polyethylene glycol (PEG) with a molecular size of 20 kDa (20K) (PEG(20K)) to obtain hbFGF derivative, PEG(20K)-hbFGF(Ser25,87,92). The optimal modified reaction was conducted at 12 degrees C for 12h with the molar ratio of PEG(20K) to hbFGF(Ser25,87,92) of 30:1. The result of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) showed that the modification rate was up to 60%.

A new eudesmane derivative, 4alpha,5alpha-dihydroxyeudesma-11(13)-en-12-oic acid, was isolated from Laggera pterodonta. Its structure was elucidated as (1) on the basis of spectroscopic evidence. Moreover, six known compounds were isolated: pterodontriol A (2), pterodontriol B, pterodonta acid (3), ilicic acid (4), costic acid and beta-amyrin.
