Mutation or deletion of the deubiquitinase USP7 causes Hao-Fountain syndrome (HAFOUS), which is characterized by speech delay, intellectual disability, and aggressive behavior and highlights important unknown roles of USP7 in the nervous system. Here, we conditionally delete USP7 in glutamatergic neurons in the mouse forebrain, triggering disease-relevant phenotypes, including sensorimotor deficits, impaired cognition, and aggressive behavior. Although USP7 deletion induces p53-dependent neuronal apoptosis, most behavioral abnormalities in USP7 conditional knockout mice persist following p53 loss.

Tertiary lymphoid structures (TLSs) are de novo ectopic lymphoid aggregates that regulate immunity in chronically inflamed tissues, including tumours. Although TLSs form due to inflammation-triggered activation of the lymphotoxin (LT)-LTβ receptor (LTβR) pathway, the inflammatory signals and cells that induce TLSs remain incompletely identified. Here we show that interleukin-33 (IL-33), the alarmin released by inflamed tissues, induces TLSs.

Conventional pelvic exenteration (PE) comprises the removal of all or most central pelvic organs and is established in clinical practise. Previously, tumours involving bone or lateral sidewall structures were deemed inoperable due to associated morbidity, mortality, and poor oncological outcomes. Recently however high-complexity PE is increasingly described and is defined as encompassing conventional PE with the additional resection of bone or pelvic sidewall structures.

Article Synopsis
  • Pseudomyxoma peritonei (PMP) is a rare condition linked to mucin-producing tumors, usually starting in the appendix, leading to mucin buildup in the abdomen.
  • Researchers conducted RNA-seq analysis on ten PMP cases and their healthy tissue to reveal 32 differently expressed genes, indicating that PMP tumors arise from goblet cells.
  • The study found significant associations between PMP tumors and important biological processes like epithelial-mesenchymal transition, angiogenesis, and inflammation, with further analysis highlighting distinct gene expressions in different PMP types, suggesting more aggressive traits in peritoneal mucinous adenocarcinomas (PMCA).

Background: Many patients who get discharged from the intensive care unit experience physical dysfunction that persists even after discharge. Physical dysfunction is associated with skeletal muscle atrophy and accompanying intensive care unit-acquired weakness in the early stages of intensive care unit admission, and early diagnosis and prevention with early mobilization are crucial. However, the amount of physical activity required for early mobilization remains controversial in critically ill patients.

Transurethral lithotripsy (TUL) using holmium YAG laser has become the standard treatment for kidney stones. Mosesᵀᴹ technology, which delivers the laser with less energy loss, has been introduced and is reported to have advantages over regular laser in terms of shorter operation time and lithotripsy efficiency, but there are few reports from general hospitals. We retrospectively compared the perioperative and postoperative outcome, and complications of 28 cases of TUL using Mosesᵀᴹ mode performed from August 2021 to January 2023 at our hospital, and 25 cases of TUL using regular laser from August 2020 to July 2021.

The gut mycobiota is crucial for intestinal homeostasis and immune function. Yet its variability and inconsistent fungal colonization of laboratory mice hinders the study of the evolutionary and immune processes that underpin commensalism. Here, we show that Kazachstania pintolopesii is a fungal commensal in wild urban and rural mice, with an exceptional ability to colonize the mouse gastrointestinal tract and dominate the gut mycobiome.
