The axolotl is broadly used in regenerative, developmental, and evolutionary biology research. Targeted gene knock-in is crucial for precision transgenesis, enabling disease modeling, visualization, tracking, and functional manipulation of specific cells or genes of interest (GOIs). Existing CRISPR/Cas9-mediated homology-independent method for gene knock-in often causes "scars/indels" at integration junctions.

Previous studies have predominantly focused on gas leakage and its subsequent toxic effects on marine organisms; however, the impacts of accidental CO leaks on carbonate dynamics during drilling and extraction remain poorly understood. In this study, we investigate carbonate parameters in two gas fields to elucidate the influences of extraction activities and natural processes on carbonate dynamics in Beibu Gulf, situated in the northern South China Sea (nSCS).

However, studies on the comprehensive effects of MPs on nitrogen cycling in crop rhizosphere soil, and the changes this effect causes to crop growth is still limited. In this study, we investigated how three types of 5 % MPs (polystyrene, PS; polyethylene, PE; polyvinyl chloride, PVC) affect soybean growth by altering rhizosphere soil nitrogen cycling.

CRISPR-mediated base editing has emerged as a powerful method for modeling diseases caused by SNVs, particularly in zebrafish, which serve as an excellent platform for investigating disease mechanisms and conducting drug screenings. However, existing cytosine base editors (CBEs) for zebrafish often have broad editing windows and strong sequence preferences, limiting their effectiveness.

Introduction: The pancreas plays a crucial role in digestion and blood glucose regulation. Current animal models, primarily mice and zebrafish, have limited the exploration of pancreatic biology from an evolutionary-developmental perspective. Tetrapod vertebrate axolotl () serves as a valuable model in developmental, regenerative, and evolutionary biology.
View Article and Find Full Text PDFAccurately defining cell boundaries for spatial transcriptomics is technically challenging. The current major approaches are nuclear staining or mathematical inference, which either exclude the cytoplasm or determine a hypothetical boundary. Here, a new method is introduced for defining cell boundaries: labeling cell membranes using genetically coded fluorescent proteins, which allows precise indexing of sequencing spots and transcripts within cells on sections.

However, several challenges remain for PRC materials in their practical application in building thermal management, including overcooling problems and unsatisfactory cooling efficiency caused by solar absorption and parasitic heat gains. In this work, fluorinated cellulose-based composite aerogels (FCCA) integrating thermal insulation and PRC were developed by a facile manufacturing strategy that combined phase separation and freeze-drying.

However, the superiority of surgery over conservative medical treatment (CMT) and the comparative benefits of different surgical procedures remain unclear. We aimed to evaluate the efficacy and safety of various surgical interventions for treating ICH.

Herein, a fluorescence/colorimetry/smartphone triple-mode sensing platform was constructed for both AChE activity monitoring and AChE inhibitor screening by exploring a Cu (I) compound, CuI (SR) (R = CHCHNH), as a fluorescent probe. In comparison of most other fluorescent probes, CuI (SR) presented exceptional stability against pH, temperature, UV irradiation, redox agents, and metal ions, as well as good recyclability due to its unique chemical structure.

Objectives: This study aimed to explore the relationship of systemic immune inflammation index (SII) and uric acid with gout using NHANES 2017-2018 data.
Method: Data were collected from the 2017-2018 circle of the NHANES database. SII was calculated based on the counts of lymphocytes (LC), neutrophils (NC), and platelets (PC).
Introduction: Amdizalisib (HMPL-689) is an ATP-competitive PI3Kδ inhibitor currently under investigation for treating Hodgkin's lymphoma. This study aimed to evaluate the metabolism, excretion, pharmacokinetics, and safety profile of amdizalisib in healthy human subjects to support its clinical application.
Methods: This Phase I clinical trial included six healthy Chinese male volunteers who received a single oral dose of 30 mg/100 µCi [C]amdizalisib suspension.

With the advancement of worldwide carbon neutralization, passive radiative cooling (PRC) has attracted tremendous interest in energy conservation by dissipating heat into the ultracold outer space without any electricity consumption. Despite some progress has been made in tailoring spectral properties for PRC, it still remains a crucial challenge in fabricating efficient and low-cost coolers for building energy saving. Herein, hierarchical cellulose-based aerogel coolers consisting of beads-on-string structural electrospun nanofibers are manufactured for all-day and all-region energy saving by combining radiative cooling and thermal insulation in one design.

However, current base editors struggle to edit cytosines in certain DNA contexts, particularly those with GC and CC pairs, limiting their use in modelling disease-related mutations. Here we show the development of zevoCDA1, an optimized cytosine base editor for zebrafish that improves editing efficiency across various DNA contexts and reduces restrictions imposed by the protospacer adjacent motif.

Objective: To explore the relationship between triglyceride-glucose index(triglyceride-glucose index) and food intake and the comprehensive compliance rate in patients with type 2 diabetes mellitus(T2DM).
Methods: A total of 6683 T2DM patients aged 18 to 75 years entered the National Metabolic Management Center(MMC) in two Yancheng hospitals from August 2020 to November 2023 were selected to analyze the relationship between T2DM standard attainment rate and relevant indicators. Univariate analysis was conducted with χ~2 test, multivariate analysis was conducted with stepwise logistic regression model, and the relationship of TyG to the overall compliance rate of T2DM was analyzed with restricted cubic spline(Restricted Cubic Spline, RCS).
Background: We concurrently developed a prospective study to assess clinical outcomes among patients receiving 9-month bedaquiline (BDQ)-containing regimens, aiming to provide valuable data on the use of this short-course regimen in China.
Methods: This open-label, randomized, controlled, multicenter, non-inferiority trial was conducted at sixteen hospitals, and enrolled participants aged 18 years and older with pulmonary rifampicin/multidrug tuberculosis. Participants were randomly assigned, in a 1:1 ratio.

Maize (Zea mays) is vital as a staple food and livestock feed crop. Yunnan is one of the main maize-producing provinces in China (National Bureau of Statistics, 2022). While corn production in Yunnan is lower than the national average, the development of drought-tolerant varieties has contributed to improving productivity.
