The axolotl is broadly used in regenerative, developmental, and evolutionary biology research. Targeted gene knock-in is crucial for precision transgenesis, enabling disease modeling, visualization, tracking, and functional manipulation of specific cells or genes of interest (GOIs). Existing CRISPR/Cas9-mediated homology-independent method for gene knock-in often causes "scars/indels" at integration junctions.

Accurately defining cell boundaries for spatial transcriptomics is technically challenging. The current major approaches are nuclear staining or mathematical inference, which either exclude the cytoplasm or determine a hypothetical boundary. Here, a new method is introduced for defining cell boundaries: labeling cell membranes using genetically coded fluorescent proteins, which allows precise indexing of sequencing spots and transcripts within cells on sections.

Passive radiative cooling (PRC) is an emerging sustainable technology that plays a key role for achieving the goal of carbon neutrality. However, several challenges remain for PRC materials in their practical application in building thermal management, including overcooling problems and unsatisfactory cooling efficiency caused by solar absorption and parasitic heat gains. In this work, fluorinated cellulose-based composite aerogels (FCCA) integrating thermal insulation and PRC were developed by a facile manufacturing strategy that combined phase separation and freeze-drying.

Acetylcholinesterase (AChE) and AChE inhibitors play critical roles in the early diagnosis and treatment of Alzheimer's disease (AD). Herein, a fluorescence/colorimetry/smartphone triple-mode sensing platform was constructed for both AChE activity monitoring and AChE inhibitor screening by exploring a Cu (I) compound, CuI (SR) (R = CHCHNH), as a fluorescent probe. In comparison of most other fluorescent probes, CuI (SR) presented exceptional stability against pH, temperature, UV irradiation, redox agents, and metal ions, as well as good recyclability due to its unique chemical structure.

With the advancement of worldwide carbon neutralization, passive radiative cooling (PRC) has attracted tremendous interest in energy conservation by dissipating heat into the ultracold outer space without any electricity consumption. Despite some progress has been made in tailoring spectral properties for PRC, it still remains a crucial challenge in fabricating efficient and low-cost coolers for building energy saving. Herein, hierarchical cellulose-based aerogel coolers consisting of beads-on-string structural electrospun nanofibers are manufactured for all-day and all-region energy saving by combining radiative cooling and thermal insulation in one design.

Cytosine base editing is a powerful tool for making precise single nucleotide changes in cells and model organisms like zebrafish, which are valuable for studying human diseases. However, current base editors struggle to edit cytosines in certain DNA contexts, particularly those with GC and CC pairs, limiting their use in modelling disease-related mutations. Here we show the development of zevoCDA1, an optimized cytosine base editor for zebrafish that improves editing efficiency across various DNA contexts and reduces restrictions imposed by the protospacer adjacent motif.

Article Synopsis
  • - Atherosclerosis leads to cardiovascular diseases due to the buildup of oxidized lipoproteins (ox-LDL) in arteries, promoting inflammation and plaque formation.
  • - Hydrogen sulfide (HS) shows promise as an anti-inflammatory agent; it reduces the uptake of ox-LDL by macrophages, while another compound, propargylglycine (PPG), increases this process.
  • - HS down-regulates receptors involved in ox-LDL uptake, specifically CD36 and SR-A, through the NF-κB signaling pathway, suggesting its potential as a therapeutic target in early atherosclerosis development.
