  • Inhibition of Notch1 signaling enhances the ability of astrocytes to generate new neurons after a stroke.
  • A rat stroke model was used to study how blocking Notch1 affects reactive astrocytes, specifically by employing the γ-secretase inhibitor DAPT at various time points post-injury.
  • Results indicated that administering DAPT 4 days after the stroke improved neurogenesis and recovery of nerve function, highlighting the significance of timing in promoting brain repair.

Article Synopsis
  • The study analyzed the levels and sources of polycyclic aromatic hydrocarbons (PAHs) in Jiaxing's river network using GC-MS, revealing significant differences between wet and dry seasons.
  • In the dry season, 10 types of PAHs were detected with higher average mass concentrations (143.83 ng/L) compared to the wet season, which had 16 types and lower concentrations (73.47 ng/L).
  • Although PAH pollution is at a low level compared to other regions, certain PAHs, particularly naphthalene and phenanthrene, pose moderate ecological risks in both seasons, indicating a need for improved pollution control measures.

  • Duchenne muscular dystrophy (DMD) and Becker muscular dystrophy (BMD) stem from mutations in the DMD gene, and this study focuses on identifying these mutations to minimize recurrence risk in affected families.
  • Researchers examined 100 unrelated DMD/BMD patients and identified pathogenic variants, including 11 novel ones, with the majority being intragenic deletions.
  • The study highlights the importance of genetic testing methods like MLPA, NGS, and HMSA and emphasizes that early genetic diagnosis can aid in prenatal options, including the termination of affected pregnancies and the successful birth of healthy children through preimplantation genetic diagnosis (PGD).

Mesenchymal stem cells (MSCs) are self-renewing multipotent cells with immunoregulatory function, which makes them attractive candidates for regenerative medicine. However, the detailed mechanisms of their immunomodulatory capacity are not fully characterized. Here, we found that casein kinase 2 interacting protein-1 (CKIP-1) expression was induced in the murine MSC cell line C3H/10T1/2 by LPS.

Adipose-derived stem cells (ASCs) are highly attractive for cell-based therapies in tissue repair and regeneration because they have multilineage differentiation capacity and are immunosuppressive. However, the detailed epigenetic mechanisms of their immunoregulatory capacity are not fully defined. In this study, we found that Mysm1 was induced in ASCs treated with inflammatory cytokines.

  • Recent technology called semiconductor sequencing platform (SSP) can help check for genetic problems before a baby is implanted in a mother's womb.
  • Scientists tested this method using DNA from IVF embryos and found it worked well with different DNA amplification tools.
  • The SSP method showed it could quickly and accurately identify genetic issues, matching results from an older method, making it a better choice for future genetic testing.

Objective: To investigate the efficacy of hematopoietic stem cells cryopreserved by ladder-style freezing from low temperature refrigerator to liquid nitrogen in treatment of hematological malignancies, and to analyze the survival condition of patients after hematopoietic stem cell transplantation.

Methods: The coyoprotectant formed by 3% hydroxyethyl starch, 4% albumin and 5% dimethyl sulfoxide (DMSO) was need for cryopreservation of hematopoietic stem cells, which were first placed in -80C low temperature refrigerator and then were stored in -196C liquid nitrogen tank. 98 cases of hemafologic malignancies (io cases of ALL, 24 cases of AML, L-cases of MM and 53 case of malignant lymphoma) were selected from January 2002 to December 2016, and recived transplantatin auto-hematopoiehc stem cells cryopresorved by above-mentined method.

It is still not clear whether Notch1 signaling inhibition can promote functional outcomes after stroke, given that it plays time-dependent roles in the sequential process of endogenous neurogenesis. The purpose of this study was to identify the appropriate time frame for Notch1 signaling inhibition according to the temporal evolution of Notch1 signaling activation and the responses of neural stem cells (NSCs), in order to target it for therapeutic intervention and stimulate neurorestorative strategies after stroke. Sprague-Dawley (SD) rats were subjected to 90-min of middle cerebral artery occlusion (MCAO).

  • cfDNA screening is a reliable and safe method for detecting fetal chromosome abnormalities, including sub-chromosomal copy number variations (CNVs), in pregnant women.
  • In a study involving over 11,000 pregnant women, 137 tested positive for aneuploidies, with a significant portion confirmed through further testing, revealing common trisomies and sex-chromosomal abnormalities.
  • The findings suggest that cfDNA screening can indicate potential parental balanced chromosome translocations, highlighting the importance of follow-up diagnostic testing.

Objective: The Notch signaling pathway is involved in angiogenesis induced by brain ischemia and can be efficiently inhibited by the -secretase inhibitor N-[N-(3,5-difluorophenacetyl)-1-alanyl]-S-phenylglycine t-butyl ester (DAPT). The aim of the present study was to noninvasively investigate the effect of DAPT treatment on angiogenesis in brain repair after stroke using magnetic resonance imaging (MRI).

Methods: Sprague-Dawley rats ( = 40) were subjected to 90 minutes of transient middle cerebral artery (MCA) occlusion and treated with PBS ( = 20) or DAPT ( = 20) at 72 hours after the onset of ischemia.

Fluid-attenuated inversion recovery (FLAIR) vascular hyperintensity (FVH) is used to assess leptomeningeal collateral circulation, but clinical outcomes of patients with FVH can be very different. The aim of the present study was to assess a FVH score and explore its relationship with clinical outcomes. Patients with acute ischemic stroke due to middle cerebral artery M1 occlusion underwent magnetic resonance imaging and were followed up at 10 days (National Institutes of Health Stroke Scale) and 90 days (modified Rankin Scale) to determine short-term clinical outcomes.

  • This study explores the use of chromosomal microarray analysis (CMA) on products of conception (POC) to identify parental balanced translocations that could contribute to infertility.
  • Over a five-year period, 1294 POC samples were analyzed, detecting subchromosomal copy number variations (CNVs) in 16 cases indicative of unbalanced translocation derivatives, all confirmed through parental genetic testing.
  • The findings highlight CMA's effectiveness in diagnosing CNVs, particularly for couples where one partner has a hidden balanced translocation without obvious chromosomal abnormalities.

Notch homolog 1 (Notch 1) signaling is regarded as a potential therapeutic target for modulating the inflammatory response and exhibiting neuroprotective effects in cerebral injury following stroke. N-[N-(3,5-difluorophenacetyl)-1-alanyl]-S-phenylglycine t‑butylester (DAPT) efficiently inhibits activation of the Notch 1 signaling pathway in microglia and may protect brain tissue from ischemic damage. However, the temporal proliferation and morphological alterations of microglia/macrophages throughout progression of the disease, as well as the comprehensive alterations of the whole brain following DAPT treatment, remain to be elucidated.
