The human DNA methyltransferases DNMT3A and DNMT3B have two types of promoters with different CpG contents.

DNA modification that is established by de novo methylation is involved in the epigenetic control of genome functions. The DNMT3A and DNMT3B genes encode putative de novo methyltransferases. In this paper, we investigated the transcriptional regulatory regions of the human DNMT3A and DNMT3B genes.

Single nucleotide polymorphisms of thrifty genes for energy metabolism: evolutionary origins and prospects for intervention to prevent obesity-related diseases.

The "thrifty" genotype and phenotype that save energy are detrimental to the health of people living in affluent societies. Individual differences in energy metabolism are caused primarily by single nucleotide polymorphisms (SNPs), some of which promote the development of obesity/type 2 diabetes mellitus. In this review, four major questions are addressed: (1) Why did regional differences in energy metabolism develop during evolution? (2) How do genes respond to starvation and affluence? (3) Which SNPs correspond to the hypothetical "thrifty genes"? (4) How can we cope with disease susceptibility caused by the "thrifty" SNPs? We examined mtDNA and genes for energy metabolism in people who live in several parts of Asia and the Pacific islands.

Sexual difference in buccal morphology of the paternal mouthbrooding cardinalfish Apogon doederleini.

The buccal morphology was compared between the sexes of the cardinalfish Apogon doederleini, in which males provide mouthbrooding. The brood size increased proportionally with male buccal space, which increased with the fourth power of the standard length. In the breeding season, males had a larger buccal space than females, whereas there was no sexual difference in the non-breeding season, suggesting sexually different flexibility in the buccal morphology.

Functional human NAIP promoter transcription regulatory elements for the NAIP and PsiNAIP genes.

Neuronal apoptosis inhibitory protein (NAIP) has been shown to inhibit apoptosis in vitro and in vivo with an expression which is regulated in a variety of cells and tissues and may be modulated by a variety of external stimuli. To understand the molecular basis of the transcriptional regulation of the NAIP gene, we have analyzed the 5'-flanking region and transcription of the human NAIP gene. The functional promoter and silencer elements were identified by luciferase reporter constructs in transient transfection experiments using four different human cells.

Syntheses and absolute stereochemistries of UPA0043 and UPA0044, cytotoxic antibiotics having a p-quinone-methide structure.

The first syntheses of new antibiotics UPA0043 and UPA0044 were accomplished starting from commercially available 18beta-glycyrrhetinic acid and vanillin. The present syntheses involve the coupling of a sesquiterpenoid aldehyde and an aryllithium, the stereoselective formation of a p-quinone-methide system, and regioselective intramolecular cyclization via an epoxy ring opening. Age-related hypermethylation of the hMLH1 promoter in gastric cancers.

To determine whether methylation of the hMLH1 promoter is related to increasing age and gastric carcinogenesis, we examined hMLH1 methylation and expression in 100 gastric cancers. hMLH1 methylation and aberrant protein expression were observed in 9 and 13 cancers, respectively. Normal and intestinal metaplastic tissues adjacent to cancers with hypermethylation did not exhibit any hMLH1 methylation, indicating that it may be specific to gastric cancers.

A gene encoding a putative GTPase regulator is mutated in familial amyotrophic lateral sclerosis 2.

Amyotrophic lateral sclerosis 2 (ALS2) is an autosomal recessive form of juvenile ALS and has been mapped to human chromosome 2q33. Here we report the identification of two independent deletion mutations linked to ALS2 in the coding exons of the new gene ALS2. These deletion mutations result in frameshifts that generate premature stop codons.

Nitrous acid interference with passive NO2 measurement methods and the impact on indoor NO2 data.

Recent research has demonstrated that nitrous acid (HONO) is produced in indoor environments by NO2 reacting with interior surfaces, and is also emitted directly by some combustion sources. We have recently characterized the interference by HONO to NO2 measurements made by several commonly used continuous NO2 monitors. This paper reports on the effect of HONO on NO2 measurements made by passive sampling devices.

Analysis of organic esters of plasticizer in indoor air by GC-MS and GC-FPD.

The aim of this study was to assess the performance of a method of analyzing organic esters of plasticizer in indoor air by sampling air in a charcoal tube and extracting the esters in toluene using a gas chromatography-mass spectrometer (GC-MS) and flame photometric detector (FPD). An internal standardization method was used for the GC-MS measurement of phthalate esters, whereas an external calibration method was employed to determine the levels of phosphate esters by FPD. The instrumental detection limit, the instrumental lower limit of determination, and the blank and method detection limits were also determined.

Malignant islet cell tumor projecting into the main pancreatic duct.

We report herein a rare case of islet cell tumor showing a unique growth pattern in a patient who developed repeated acute pancreatitis as the tumor's initial symptom. Preoperative imaging examinations showed dilatation of the main pancreatic duct (MPD) and cysts around the pancreatic tail. A distal pancreatectomy with splenectomy was performed because the pancreatitis was localized in the distal pancreas and was not controlled by various drug therapies.

Monogamy and sex role reversal in the pipefish Corythoichthys haematopterus.

We observed the mating pattern and social behaviour of the pipefish Corythoichthys haematopterus in temperate waters of Japan during three successive breeding seasons. Males cared for a clutch in their brood pouch for 9-19 days until hatching and had several broods in the season with nonbrooding intervals of only 1 or 2 days. The population sex ratio was female biased and some females were always excluded from reproduction.

The autosomal recessive form of juvenile amyotrophic lateral sclerosis (ALS2) has previously been mapped to the 1.7-cM interval flanked by D2S116 and D2S2237 on human chromosome 2q33-q34.

Characteristics of sorption losses of polychlorinated biphenyl congeners onto glass surfaces.

Sorption losses to glass surfaces of five polychlorinated biphenyl (PCB) congeners in aqueous solutions were investigated. Adsorption/desorption experiments were conducted under conditions that simulated actual sample handling procedures for environmental samples. It was found that the adsorption loss is related to the degree of chlorination.

Interpretation and enantiomer analysis of methamphetamine abusers' urine and illegally brewed methamphetamine crystals.

This study deals with the high-performance liquid chromatographic identification of methamphetamine (MAMP) and amphetamine (AMP) enantiomers (d- and l-forms) in five illicit MAMP crystals and in urine specimens from 30 Japanese MAMP abusers. The analysis revealed that two of the types of crystals have a different optical purity ratio (l/d) and the other three have a single crystal of either the d- or l-enantiomer. The l/d ratios of two types of crystals were 0.

Methylation of the hMLH1 promoter in familial gastric cancer with microsatellite instability.

Microsatellite instability (MSI), which is recognized as an important mechanism in tumorigenesis, has been reported in familial gastric cancers (FGC). However, genetic defects responsible for this phenotype, that is, mutations in mismatch-repair genes such as hMLH1 and hMSH2, have not been detected in most FGC cases. Earlier studies have shown that the promoter region of the hMLH1 gene was methylated in some sporadic colorectal and endometrial cancers.

A core promoter and a frequent single-nucleotide polymorphism of the mismatch repair gene hMLH1.

The hMLH1 gene encodes a protein that is involved in the DNA mismatch repair system. The coding region of the hMLH1 gene has been known to be mutated in a subset of patients with hereditary nonpolyposis colorectal cancer (HNPCC). Our current research characterized the promoter region of the hMLH1 gene and searched for mutations correlating to HNPCC.

Two short sequences have positive effects on the human p27Kip1 gene transcription.

The cyclin-dependent kinase (Cdk) inhibitor p27Kip1 plays an important role in the progression from G1 to S phase in the cell cycle. To study the activities of its promoter and other regulatory elements, we have cloned and characterized the 5'-flanking region of the human p27Kip1 gene. This region, about 3kb in length, is GC-rich and shares homology with that of the mouse p27Kip1 gene.

Impact of residential nitrogen dioxide exposure on personal exposure: an international study.

Nitrogen dioxide (NO2) concentrations were measured during two-day winter periods in indoor and outdoor environments, and these concentrations were compared with simultaneously measured personal exposures in 18 cities in 15 countries around the world. Information was also gathered on activity patterns and household characteristics in order to determine the influences of these factors on personal exposures. All NO2 measurements were taken using passive filter badges.

Residual Perchloroethylene in Dry-Cleaned Acetate: The Effect of Pressing and Extent of Inter-Dry-Cleaner Variability.

Measurements of residual perchloroethylene (PCE), a dry-cleaning solvent associated with human health effects, were made in dry-cleaned acetate cloth to enable improved characterizations of both occupational and environmental exposure. A limited sample size (25 acetate cloths) was used to explore the extent of inter-dry-cleaner variability in residual PCE and to characterize the effect of the pressing operation on residual PCE. A new method, which uses carbon-disulfide as the direct extracting agent, proved effective in the analysis of residual PCE, with a recovery-efficiency ≈ 75%.

Determinants of nitrogen dioxide concentrations in indoor ice skating rinks.

Objectives: The combination of poor ventilation and fuel-powered ice resurfacers has resulted in elevated nitrogen dioxide (NO2) concentrations in many indoor ice skating rinks. This study examined the factors influencing concentrations and the effects of various engineering controls in ice rinks with different resurfacer fuels.

Methods: Indoor NO2 concentrations were measured in 19 enclosed ice skating rinks over 3 winters by means of passive samplers, with 1-week average measurements during the first winter pilot study and single-day working-hour measurements in the final 2 winters.

Association equilibrium of d-methamphetamine and l-methamphetamine with serum albumin.

The association equilibria for complex formation between serum albumin (bovine, rat) and the optical isomers of methamphetamine (MAMP) was determined using an ultrafiltration method. It was found that serum albumin/d-MAMP and serum albumin/l-MAMP complexes had distinctly different Scatchard plots with bovine and rat albumin. The binding parameters of each association equilibrium were estimated from the Scatchard plots by Rosenthal's graphic method.

The Boston residential nitrogen dioxide characterization study: classification and prediction of indoor NO2 exposure.

Many epidemiological studies have used house characteristics associated with indoor sources as simplified proxies for personal nitrogen dioxide (NO2) exposure. Stove type and presence of a pilot light often been used as the two key characteristics, but significant overlaps have remained in the NO2 concentrations in the exposed and unexposed groups. This has contributed to inconsistencies in epidemiological findings, due to potential misclassification of exposure.
