Enduring challenges for quantitative analysis in nanopore sensing are the detection of low biomarker concentrations in reasonable time frames and the detection of multiple biomarkers in the same sample. Herein we report an optical blockade nanopore sensor strategy that can detect more than a protein at femtomolar concentrations in rapid time (approximately 12 min). This is done using a nanopore array functionalized with an aptamer that can bind to two different but related target proteins.

Background: We aim to determine the effectiveness of a community-based, health instructor led, multifaceted family intervention, as compared with usual care, on blood pressure (BP) management among Chinese rural residents, with or without hypertension.
Methods/design: The Healthy Family Program is a cluster randomized controlled trial being undertaken in 80 villages (each with approximately 100 residents) with a target to enroll a total of 8000 older adults (aged 40-80 years). Villages were randomly assigned in a 1:1 ratio to either an intervention group to receive multifaceted strategies or a control group to continue with usual standard of care.
Fluorescent probes for in vivo hypochlorous acid (HClO) imaging often face challenges of low selectivity and high cytotoxicity, largely due to poor analyte recognition and water-insoluble aromatic skeletons. To address this, we synthesized fluorescein hydrazide by introducing a spiro-lactam unit into fluorescein, which offers high emission intensity and molar absorption. The five-membered heterocycle in fluorescein hydrazide is selectively disrupted by HClO, enhancing the conjugated system and electron delocalization of the fluorophore, resulting in highly sensitive fluorescence detection of HClO.

Methods: A total of 70 GBC patients, 70 patients with benign gallbladder diseases (gallbladder stones and gallbladder polyps), and 70 normal health examination people were selected as the malignant, benign, and normal groups, respectively. The differences in serum levels and positive rates of PIVKA-II and CA125 were compared.
Copper (cu) is an essential micronutrient required for numerous metabolic processes. It plays a crucial role in cellular respiration by participating in the electron transport chain and facilitating numerous biological reactions. Various diseases, including cancer, demonstrate localized elevation of copper levels and/or alterations in the overall distribution of copper.

Methods: ALF patients (n = 120, ALF group) and 106 healthy subjects (control group) were enrolled. Serum was collected to detect alanine aminotransferase (ALT), aspartate aminotransferase (AST), and total bilirubin (TBIL) by automatic biochemical analyzer, and miR-130a by real-time fluorescence quantitative PCR.
A competitive electrochemiluminescence (ECL) immunosensor is proposed to accurately and rapidly assess okadaic acid (OA) levels in shellfish using a novel self-reinforced solid-state ECL marker, which is essential for ensuring seafood safety. Graphene quantum dots doped with nitrogen and sulfur (N,S-GQDs) were synthesized, for the first time, through the electrolysis of graphite in 3-(N-morpholine) propane sulfonic acid solution. Intriguingly, these N,S-GQDs exhibited exceptional co-reactant properties, significantly enhancing the anodic ECL performance of Ru(bpy) in a phosphate-buffered saline solution.

This study presents the development of a dual-mode aptasensor for the sensitive detection of kanamycin (KAN), utilizing both fluorescence and colorimetric signals. The aptasensor was constructed using amino-functionalized silica nanoparticles (SiO) combined with copper nanoclusters (CuNCs) and DNA-templated silver nanoclusters (DNA-AgNCs). Encapsulating CuNCs within SiO (CuNCs@SiO) enhanced their stability by shielding them from environmental interference, while maintaining their bright blue fluorescence as a reference signal.

Methods: Data obtained from liver cancer screening in 2016-2018 and 2019-2020 in patients with hepatocellular carcinoma in Xiamen were collected using Xiamen Healthcare Big Data Platform. Epidemiological characteristics were analyzed descriptively by different years.
Systematic construction of isoreticular hydrogen-bonded organic frameworks (HOFs) promises tailored material properties crucial for diverse applications, yet is challenging due to the weak, flexible, and non-directional nature of hydrogen bonds. Herein, we develop an "integrated monomer synthesis-framework assembly" (ISA) methodology for constructing a series of isoreticular HOFs. Unlike traditional methods where monomers are first synthesized and then assembled into HOFs, the ISA system employs dicyandiamide rigid hydrogen-bonded hexameric clusters as connecting nodes to covalently react with planarized C-symmetric cyano-precursors (C-CPs) to generate diaminotriazine (DAT) monomers, while simultaneously inducing the directional assembly into isoreticular (6,3)-net hcb topological DAT-C-HOFs.

In this study, the microbial communities and volatile compounds were analyzed and compared during the storage of Daqu. Virgibacillus, Bacillus and Kroppenstedtia were the dominant bacterial genera, and Thermoascus, Thermomyces and Aspergillus were the dominant fungal genera.

TBK1 is an important IFN antiviral signalling factor, and in previous work black carp TBK1 (bcTBK1) and black carp IRF5 (bcIRF5) together promoted cell death in GCRV-infected cells. In this research, bcTBK1 and bcIRF5 were investigated both in vivo and in vitro to delineate their individual and combined functions. This study demonstrated that both bcTBK1 and bcIRF5 expressions were modulated in response to GCRV infection across the intestine, gill, kidney and spleen.

Signal Recognition Particle 54 kDa (SRP54) is a subunit of the signal recognition particle (SRP), a cytoplasmic ribonucleoprotein complex guiding the transportation of newly synthesized proteins from polyribosomes to endoplasmic reticulum. In mammals, it has been reported to regulate the RLR signaling pathway negatively by impairing the association between MAVS and MDA5/RIG-I. However, the role of SRP54 in teleost antiviral innate immune response remains obscure.

Simple and fast quantification of UDG activity is essential for biological assay and clinical diagnosis, since its aberrant level is associated with DNA damage and various diseases. Herein, we developed a fully integrated "sample in-signal out" distance-based paper analytical device (dPAD) for visual quantification of UDG using a flow-controlled uracil-rich DNA hydrogel (URDH).

Drug therapy is the core treatment strategy for leukemia, but the current therapeutic drugs have defects such as low bioavailability, large adverse reactions and inconvenient intravenous administration. Targeted therapy can combine drugs with specific carcinogenic sites on cells to kill cancer cells and avoid damage to normal cells, which has gradually become the mainstream method of leukemia treatment.

Influenza (flu) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) exhibit similar clinical symptoms, complicating the diagnosis and clinical management of these critical respiratory infections. Thus, there is an urgent need for rapid on-site detection technologies that can simultaneously detect SARS-CoV-2 and influenza A viruses. Here, we have developed the first platform that combines in situ sampling with immune swabs and multichannel surface-enhanced Raman spectroscopy (SERS) for simultaneous screening of these two respiratory viruses in a single assay.
