HER2 antagonists remain the cornerstone of therapy for patients with HER2-positive breast cancer. This study introduces a novel small-molecule inhibitor of DNA methyltransferase 1 (DNMT-1), referred to as DI-1, designed to synergize with HER2 antagonists in treating HER2-positive breast cancer cells. Clinical data reveal a negative correlation between DNMT-1 expression and PTEN levels, and a positive correlation with the methylation rates of PTEN's promoter.

5' untranslated regions (UTRs) are important sequence elements that modulate the expression of genes. Using the β-glucuronidase (GUS) reporter gene driven by the GluC promoter for the rice-seed storage-protein glutelin, we evaluated the potential of the 5'-UTRs of six seed storage-protein genes in enhancing the expression levels of the foreign gene in stable transgenic rice lines. All of the 5'-UTRs significantly enhanced the expression level of the GluC promoter without altering its expression pattern.

Article Synopsis
  • - The study addresses the lack of effective endosperm-specific promoters needed to express recombinant proteins in cereal crops, particularly focusing on different seed storage glutelin genes from rice.
  • - Six rice glutelin gene promoters (GluA-1, GluA-2, GluA-3, GluB-3, GluB-5, and GluC) were analyzed for their gene expression patterns using beta-glucuronidase (GUS) as a reporter, revealing distinct expression areas within the endosperm.
  • - Among the promoters, GluC exhibited the highest activity during seed maturation and lacked certain motifs associated with endosperm specificity, indicating that additional regulatory mechanisms may play a role

Lignin, one of the main components in vascular plants, is important for the adaptation of terrestrial plants to environment during evolution. However, its presence in plants has negative effects on wood processing during pulping and stock breeding. Therefore much attention has been focused on the regulation of lignin biosynthesis.
