Background: Exercise, especially high-intensity interval training (HIIT), can increase mitochondrial respiratory capacity and enhance muscular endurance, but its systemic burden makes it difficult to safely and continuously prescribe for patients with chronic kidney disease (CKD)-related cachexia who are in poor general condition. In this study, we examined whether HIIT using electrical stimulation (ES), which does not require whole-body exercise, improves muscle endurance in the skeletal muscle of 5/6 nephrectomized rats, a widely used animal model for CKD-related cachexia.

Methods: Male Wistar rats (10 weeks old) were randomly assigned to a group of sham-operated (Sham) rats and a group of 5/6 nephrectomy (Nx) rats.

We describe a case of a 47-year-old male patient with initially unresectable intrahepatic cholangiocarcinoma of the right liver lobe with tumor thrombi extending from the right bile duct to the common and left bile ducts. Conventional chemotherapy with gemcitabine and cisplatin for 19 months resulted in progressive disease. Subsequently, a comprehensive genome profile revealed fibroblast growth factor receptor 2 rearrangement, and hence, pemigatinib administration was initiated.

β-Aspartyl compounds, such as β-aspartyl hydroxamate (serine racemase inhibitor), β-aspartyl-l-lysine (moisture retention), and β-aspartyl-l-tryptophan (immunomodulator) are physiologically active compounds. There is limited literature on the development of effective methods of production of β-aspartyl compounds. In this study, we describe the biochemical characterization of asparagine synthetase (AS) from Streptococcus thermophilus NBRC 13957 (StAS) and the enzymatic synthesis of β-aspartyl compounds using StAS.

Urothelial carcinoma of the bladder with osteoclast-like giant cells (UCOGCs) is rare among the subtypes of poorly differentiated urothelial carcinoma. Its clinical significance and optimal treatment are unknown, and few reports on genomic analysis of UCOGCs have been reported. Detailed analysis including genetic analysis for rare type variants of cancer could be a foothold for further research.

Fear and anxiety among patients are sometimes evoked in dental clinics due to the sound of dental drills. This study aimed to explore the impact of age-related hearing loss in the extended high frequency (EHF) range above 8 kHz on individuals' subjective discomfort towards dental drill noise. After measuring pure-tone audiometric thresholds at both conventional and extended high frequencies, we used a psychoacoustic approach to evaluate subjective impressions of four dental drill sound stimuli, which featured varying frequency components, in 62 participants (aged 12-67 years).

Background: POCUS training courses are effective at improving knowledge and skills, but few studies have followed learners longitudinally post-course to understand facilitators, barriers, and changes in POCUS use in clinical practice. We conducted a prospective observational study of physicians who attended 11 standardized POCUS training courses between 2017 and 2019 in Japan. Physicians who attended a standardized POCUS course were surveyed about their current frequency of POCUS use of the heart, lung, abdomen, and lower extremity veins, and perceived barriers and facilitators to POCUS use in clinical practice.

  • Advanced pancreatic ductal adenocarcinoma (PDAC) has different subtypes, with the basal-like subtype showing worse prognosis and chemotherapy resistance compared to the classical subtype.
  • This study evaluated immunohistochemistry (IHC) staining of four markers on biopsy samples from 190 untreated advanced PDAC patients, identifying three distinct patterns: Classical, Transitional, and Basal-like.
  • The Basal-like pattern was linked to poorer survival rates and associated with specific histological features, making IHC expression patterns a valuable tool for predicting prognosis and guiding treatment decisions in advanced PDAC.

  • A new model for predicting germline pathogenic variants (GPVs) of the APC gene in patients with adenomatous polyposis is being developed to aid in clinical diagnosis and management, especially when genetic testing is unavailable.
  • In a study of 162 patients, 55.6% were found to have GPVs of the APC gene, with key predictors identified, including age under 40, 100 or more polyps, fundic gland polyposis, and a family history of colorectal polyposis.
  • The predictive model demonstrated high accuracy (area under the curve of 0.91) and aims to assist both patients and healthcare providers in deciding on the necessity of genetic testing.

Cases of relatively safe dental implant treatment in patients with low-volume bisphosphonate (BP) have been gradually reported. Although bone augmentation is commonly used when the bone volume is insufficient for implant placement, the studies and case reports regarding the safety of bone augmentation in patients treated with BP remain insufficient. Herein, we report a case wherein bone augmentation was performed after BP treatment with bone healing realized according to imaging, and we review the literature regarding BP and bone augmentation.

The development of antifungal drugs requires novel molecular targets due to limited treatment options and drug resistance. Through chemical screening and establishment of a novel genetic technique to repress gene expression in , the primary causal fungus of dermatophytosis, we demonstrated that fungal Cdc42 and Rac GTPases are promising antifungal drug targets. Chemical inhibitors of these GTPases impair hyphal formation, which is crucial for growth and virulence in .

Article Synopsis
  • The study aimed to assess how changes in hemoglobin (Hb) levels from discharge to one year later affect outcomes for patients with heart failure and preserved ejection fraction (HFpEF), using a lower Hb cut-off than standard WHO guidelines.
  • Researchers analyzed 547 HFpEF cases, categorizing them based on Hb levels at discharge and one year post-discharge to determine the incidence of major adverse cardiovascular events (MACE).
  • Findings indicated that patients whose Hb worsened (G3) had significantly higher rates of MACE compared to those with stable higher Hb levels (G4), while those who improved their anemia (G2) showed no significant difference in MACE compared to G4.
