  • - Dermatophagoides farinae is a common indoor allergen that has a strong ability to withstand temperature changes, yet the specifics of this tolerance mechanism are not well understood.
  • - Researchers aimed to identify the differentially expressed genes (DEGs) related to temperature stress using qRT-PCR but faced challenges due to the lack of stable reference genes.

Autophagy is crucial for immune defense against (Mtb) infection. Mtb can evade host immune attack and survival within macrophages by manipulating the autophagic process. MicroRNAs (miRNAs) are small, non-coding RNAs that are involved in regulating vital genes during Mtb infection.

  • Alcohol dependence is a major global health issue, with new research highlighting the influence of gut microbiota on brain function and addiction, in addition to traditional views on neurotransmitters.
  • A study using a rat model found that chronic alcohol consumption primarily altered the composition of gut microbiota in the colon, rather than its diversity or richness in the jejunum and colon.
  • Changes in specific bacterial genera and increased functions related to amino acid metabolism due to alcohol abuse suggest that targeting gut microbiota could offer new treatment strategies for alcohol addiction.

Anhui Province has been one of typical epidemic areas of schistosomiasis in East China as a wide range of large lake and marshland regions provide an ideal environment for growth and reproduction of the intermediate snail host. With the completion of the Yangtze River-Huaihe River Water Transfer Project (YHWTP), launched by the end of 2016, the epidemic areas are expected to expand and controlling schistosomiasis remains a challenge. Based on annual surveillance data at the county level in Anhui for the period 2006-2015, spatial and temporal cluster analyses were conducted to assess the pattern of risk through spatial (Local Moran's I and flexible scan statistic) and space-time scan statistic (Kulldorff).

The role of bacteria is unclear in the facial skin lesions caused by Demodex. To shed some light on this issue, we conducted a case-control study comparing cases with facial dermatoses with controls with healthy skin using denaturing gradient gel electrophoresis (DGGE) technique. The bacterial diversity, composition, and principal component were analyzed for Demodex bacteria and the matched facial skin bacteria.

[Human Demodicosis: Emerging Dermatosis Caused by Demodex].

Zhongguo Ji Sheng Chong Xue Yu Ji Sheng Chong Bing Za Zhi

October 2016

Article Synopsis
  • Demodex mites, discovered 175 years ago, raise questions about their role in skin diseases due to high infection rates but low cases of demodicosis, leading to confusion in clinical evaluations.
  • Recent concerns have emerged over the pathogenic effects of Demodex mites, especially in relation to skin conditions like rosacea, but many other lesions linked to the mites are often misdiagnosed.
  • This review aims to provide a comprehensive overview of Demodex, including its history, typical cases, pathogenesis, and treatment options, in order to increase awareness and understanding of demodicosis in society.

The transcriptomic data of Sarcoptes is still lacking in the public database due to the difficulty in extracting high-quality RNA from tiny mites with thick chitin. In this study, total RNA was extracted from live Sarcoptes mites for quality assessment, RNA-Seq, functional annotation, and coding region (CD) prediction and verification. The results showed that the sample JMQ-lngm was qualified for cDNA library construction.

To solve the long-existing difficult problems in extracting RNA and constructing a complementary DNA (cDNA) library for trace mites, we conducted a further comparative experiment among three RNA extraction methods (TRIzol method, Omega method, and Azanno method) based on our previous attempts at the construction of cDNA library of mites, with Psoroptes cuniculi still used as the experimental subject. By subsequently decreasing the number of mites, the least number of mites needed for RNA extraction of each method were found by criteria of completeness, concentration, and purity of the extracted RNA. Specific primers were designed according to the allergen Pso c1, Pso c2, and Actin gene sequences of Psoroptes to test the reliability of cDNA library.

RNA extraction and construction of complementary DNA (cDNA) library for mites have been quite challenging due to difficulties in acquiring tiny living mites and breaking their hard chitin. The present study is to explore a better method to construct cDNA library for mites that will lay the foundation on transcriptome and molecular pathogenesis research. We selected Psoroptes cuniculi as an experimental subject and took the following steps to construct and verify cDNA library.

  • * Researchers extracted gDNA from Psoroptes and Otodectes mites and analyzed rDNA ITS2 and mtDNA COI sequences to improve classification, finding rDNA ITS2 more effective for distinguishing between genera and species.
  • * The study revealed that while genetic distinctions based on host species were significant, geographical isolation did not play a role in differentiating certain mite species that parasitize similar animals like dogs and cats.

Article Synopsis
  • - The study investigates the species classification of Sarcoptes mites that infect humans (S. hominis) and dogs (S. canis) in China by analyzing genetic material from both types of mites.
  • - Analysis of mitochondrial DNA (mtDNA) revealed five distinct groups within the Sarcoptes family, identifying four variants of S. hominis and one for S. canis, while showing that S. canis mites from different regions share closer genetic relationships.
  • - The findings indicate that the 317-bp mtDNA cox1 sequence serves as a reliable DNA barcode for accurately identifying Sarcoptes mites, highlighting geographical differences among variants of S. hominis but not among S. canis.

Traditional classification of Demodex mites by hosts and phenotypic characteristics is defective because of environmental influences. In this study, we proposed molecular identification of four phenotypes of two human Demodex species based on mitochondrial cox1 fragments for the first time. Mites collected from sufferers' facial skin were classified into four phenotypes: phenotype A-C with finger-like terminus, and phenotype D with cone-like terminus.

For a long time, classification of Demodex mites has been based mainly on their hosts and phenotypic characteristics. A new subspecies of Demodex folliculorum has been proposed, but not confirmed. Here, cox1 partial sequences of nine isolates of three Demodex species from two geographical sources (China and Spain) were studied to conduct molecular identification of D.
