Objective: To detect the gene expression profile in gastric cancer cell cycle and explain the mechanism of gastric cancer cell proliferation by a genomic study.

Methods: Gastric cancer cells MKN45 were synchronized at G2/M and G1/S point by nocodazole-thymidine and double thymidine methods. The synchronizing degree of cells was monitored by flow cytometry.

Article Synopsis
  • The study aimed to investigate how depleting the polo-like kinase 1 (plk1) gene affects the growth of the MKN45 gastric cancer cell line both in the lab (in vitro) and in live animals (in vivo).
  • Researchers used RNA interference to reduce plk1 expression, measuring its effects on cell cycle, apoptosis, and proliferation through various methods including quantitative PCR and flow cytometry.
  • Results showed that targeting plk1 with siRNA led to decreased cell proliferation and increased apoptosis in MKN45 cells, suggesting plk1 is a promising new target for treating gastric cancer.

Objective: To observe the effect of inhibition of polo like kinase1 (plk1) gene expression on apoptosis induction in gastric cancer cell line-MKN45 and discuss the vital role of plk1 proliferation and viability of gastric cancer cells.

Methods: The plk1 expression was inhibited by chemically synthesized siRNA. The plk1 mRNA and protein level were respectively measured by real-time quantitative PCR and Western blotting.
