Article Synopsis
  • Extramammary Paget's disease (EMPD) is a rare skin cancer with an unclear cause and potential to spread, where surgery is the main treatment but more effective therapies are needed.
  • In a study analyzing 815 EMPD patients, factors like older age, metastasis, and radiotherapy were linked to worse outcomes, while chemotherapy seemed to have a negative impact on patient prognosis.
  • The research indicated that EMPD tissues had more immune-related activity, particularly in patients with low ERBB2 expression, suggesting that these patients might benefit from immunotherapy focusing on B cells.

The healing of large skin defects remains a significant challenge in clinical settings. The lack of epidermal sources, such as autologous skin grafting, limits full-thickness skin defect repair and leads to excessive scar formation. Skin organoids have the potential to generate a complete skin layer, supporting in-situ skin regeneration in the defect area.

Most deaths in breast cancer patients are attributed to metastasis, and lung metastasis is associated with a particularly poor prognosis; therefore it is imperative to identify potential target for intervention. The transforming growth factor-β (TGF-β) pathway plays a vital role in breast cancer metastasis, in which Smad3 is the key mediator and performs specific functions by binding with different cofactors. However, Smad3 cofactors involved in lung metastasis have not yet been identified.

  • p53 is a crucial tumor suppressor, and disruptions in its function can lead to tumor development, with previous research highlighting that certain Krüppel-associated box (KRAB) domain zinc-finger proteins (KZFPs) interact with and regulate p53 activity.
  • Recent analysis of gastrointestinal cancers identified ZNF8, a protein that interacts with p53, as a negative prognostic indicator for patients, as it reduces p53's transcriptional activity in colon cancer cells.
  • Experiments showed that manipulating ZNF8 levels affects cancer cell behaviors like growth and metastasis, and its higher expression in cancer tissues correlates with poorer patient survival, suggesting ZNF8 could be a

Introduction: The tumor microenvironment of hepatocellular carcinoma is composed of multiple cells, and the interactive communication between cells drives tumor progression and characterizes the tumor. Communication between cells is mainly achieved through signal transduction between receptor ligands, and the rise of single-cell technology has made it possible to analyze the communication network between cells.

Methods: We applied a train of bioinformatic techniques and in vitro experiments.

Fibroblast growth factor receptor 2 (FGFR2) is frequently activated by overexpression or mutation, and an abnormal fibroblast growth factor (FGF)/FGFR signaling pathway is associated with the occurrence, development, and poor prognosis of colorectal cancer (CRC). Our preliminary analysis found that plasminogen activator inhibitor-1 (PAI-1) expression may be related to FGF/FGFR signaling, however, their role in the tumor immune microenvironment remains unclear. In this study, we observed markedly higher PAI-1 expression in CRC patients with poor survival rates.
