Background: The cGAS-STING pathway is an important component of the innate immune system and plays significant role in acetaminophen-induced liver injury (AILI). Pentagalloylglucose (PGG) is a natural polyphenolic compound with various beneficial effects, including anti-cancer, antioxidant, anti-inflammatory, and liver-protective properties; however, whether it can be used for the treatment of AILI and the specific mechanism remain unclear.
Materials And Methods: A cell culture model was created to study the effect of PGG on cGAS-STING pathway activation using various techniques including western blotting (WB), real-time quantitative polymerase chain reaction (RT-qPCR), immunofluorescence (IF), and immunoprecipitation (IP).
Background: Nonalcoholic fatty liver disease (NAFLD) is a prevalent chronic liver ailment that can lead to serious conditions such as cirrhosis and hepatocellular carcinoma. Hepatic Nogo-B regulates glucose and lipid metabolism, and its inhibition has been shown to be protective against metabolic syndrome. Increasing evidence suggests that imbalances in the gut microbiota (GM) and lipid metabolism disorders are significant contributors to NAFLD progression.

Hence, targeted inhibition of the activation of the cGAS-STING pathway is potentially valuable in the treatment of disease. The primary active component of Salvia miltiorrhiza is total tanshinone (TTN).

The study established an inflammasome activation model of primed bone marrow-derived macrophages(BMDMs), and its molecular mechanism was investigated by Western blot(WB), immunofluorescence staining, enzyme-linked immunosorbent assay(ELISA), and flow cytometry. C57BL/6J mice were randomly divided into a blank control group, a TGP group, a model group(LPS group), LPS+low-and high-dose TGP groups, LPS+MCC950 group, and LPS+MCC950+TGP group, with eight mice per group.

Liuwei Wuling tablet (LWWL) is a traditional Chinese medicine formula, with a protecting liver and decreasing enzyme activity, usually used to treat chronic hepatitis B with NAs in clinic. However, its main active ingredients and mechanism of action have not been fully investigated.

Epimedium flavonoid (EF), a major active ingredient isolated from the medicinal plant Epimedium, has been reported to have good anti-inflammatory activity, but its exact mechanism of action remains unclear. In the present study, we found that EF in mouse bone marrow-derived macrophages (BMDMs), THP-1 (Tohoku Hospital Pediatrics-1) as well as in human peripheral blood mononuclear cells (hPBMC) inhibited the activation of the cGAS-STING signalling pathway, which subsequently led to a decrease in the expression of type I interferon (IFN-β, CXCL10 and ISG15) and pro-inflammatory cytokines (IL-6 and TNF-α).

Here, we further develop the application of TPE-CNCs for fluorescence detection of Fe in aqueous solutions. The fluorescence of TPE-CNC aqueous suspensions is rapidly quenched (response time less than 10 s) due to the electron-transfer process between TPE and Fe upon addition of Fe.

In this study, we have innovatively proposed a method for surface modification of ultra-high molecular weight polyethylene (UHMWPE) artificial joint materials with graphene oxide (GO) infiltrated into UHMWPE substrate by ultrasonic induction. The mechanical properties of UHMWPE nanocomposites with GO infiltrated by ultrasonic induction were compared with that of GO mixed. The molecular structure, wettability, peak load, and bio-tribological behavior of GO/UHMWPE nanocomposites were studied using fourier transform infrared spectroscopy, contact angle measuring instrument, electronic universal material testing machine, tribometer, and profilometer, respectively.

In this work, an aggregation-induced emission (AIE) sensor for the detection of Fe ions was fabricated through the electrostatic interaction between 1,1,2-triphenyl-2-[4-(3-sulfonatopropoxyl)-phenyl]-ethene sodium salt (SPOTPE) and quaternized cellulose (QC). The structure and properties of the SPOTPE/QC nanocomplex were studied by using ¹H NMR, spectrofluorophotometer, transmission electron microscopy (TEM), and dynamic laser light scattering (DLS). An aqueous solution of SPOTPE and QC resulted in a remarkably enhanced cyan fluorescence in comparison to that of the SPOTPE solution.
