Topical absorbable hemostats are routinely utilized in surgical procedures to assist in controlling intraoperative bleeding. SURGICEL Original Absorbable Hemostat, one of the most frequently used adjunctive hemostats, is composed of oxidized regenerated cellulose (ORC). We report here that a novel powdered form of ORC, composed of aggregates of ORC fine fibers, provides additional valuable hemostatic performance characteristics and retains the biochemical and bactericidal profile of the parent ORC fabric.

Methods: Test and control sutures (3-4 cm) were implanted subcutaneously in the dorsal-lateral regions (control on the left side, test on the right side, approximately 3-5 cm apart) in 10 female Hartley guinea pigs (300-400 g) and 10 Swiss Webster mice (20-35 g) via a 20-gauge catheter. The test material was poliglecaprone 25 suture with triclosan (2-0, undyed), and the control material was poliglecaprone 25 suture (2-0, undyed).

Methods: Poliglecaprone 25 sutures with and without triclosan were tested for in vitro efficacy against Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), Staphylococcus epidermidis, methicillin-resistant Staphylococcus epidermidis (MRSE), Klebsiella pneumoniae, and Escherichia coli by a zone of inhibition assay. The suture also was tested against Escherichia coli in a colonization assay in a dynamic model simulating in vivo conditions.
A nisin-resistant mutant of Listeria monocytogenes Scott A has been characterized by comparing its phospholipid composition with the nisin-sensitive parental strain. The total phospholipids of resistant cells were significantly ( < 0.001) decreased compared to the parental strain.

In brain heart infusion medium, spontaneous nisin resistance frequencies were in the range of 10 to 10 when exposed to nisin at concentrations 2 to 8 times the minimal inhibitory concentrations. A resistant mutant of Listeria monocytogenes Scott A (2000 U nisin per ml) was obtained by increasing stepwise exposure to nisin and was subsequently characterized.
