Developing methodologies for the expedient construction of biologically important δ-valerolactones bearing a privileged azaarene moiety and a sterically congested all-carbon quaternary stereocenter is important and full of challenges. We present herein a novel multicatalytic strategy for the stereodivergent synthesis of highly functionalized chiral δ-valerolactones bearing 1,4-nonadjacent quaternary/tertiary stereocenters by orthogonally merging borrowing hydrogen and Michael addition between α-azaaryl acetates and allylic alcohols followed by lactonization in a one-pot manner. Enabled by Cu/Ru relay catalysis, this cascade protocol offers the advantages of atom/step economy, redox-neutrality, mild reaction conditions, and broad substrate tolerance.

Article Synopsis
  • The study investigates the link between urticaria (a skin condition) and mental disorders, confirming that individuals with urticaria are at a higher risk for conditions like ADHD and schizophrenia.
  • Using data from various genome studies, researchers found that different types of urticaria are associated with specific mental health risks and examined the role of inflammatory cytokines.
  • The findings suggest that inflammatory cytokines do not mediate the relationship between urticaria and mental disorders, indicating a need for integrated treatment approaches that consider both physical and mental health.

Article Synopsis
  • This study explored the potential causal link between allergic diseases and dementia using advanced statistical methods called Mendelian randomization, analyzing data from thousands of patients.
  • The results showed that conditions like atopic dermatitis and asthma may reduce the risk of certain types of dementia, while allergic rhinitis appeared to increase the risk for Alzheimer's disease and vascular dementia.
  • Overall, the findings suggest that ADHD is more clearly linked with increased dementia risk, and other allergic conditions may be affected by confounding factors rather than having a direct causal relationship.

Transition-metal catalyzed functionalization of ACPs has been widely investigated in cycloaddition and 1,3-difunctionalization reactions. However, the transition metal catalyzed nucleophilic reactions of ACPs have rarely been reported. In this article, an enantio-, site- and /-selective addition of ACPs with imines for the synthesis of dienyl substituted amines has been developed palladium- and Brønsted acid co-catalysis.
