Proteolysis targeting chimeras (PROTACs) hold immense promise for targeted protein degradation; however, challenges such as off-target effects, poor drug-likeness properties, and the "hook effect" remain. This study introduces Nano-Click-formed PROTACs (Nano-CLIPTACs) for precise tumor protein degradation in vivo. Traditional PROTACs with high molecular weight were first divided into two smaller druglike precursors capable of self-assembling to form functional PROTACs through a bioorthogonal reaction.

Background/objectives: Clinical use of poly(ADP-ribose) polymerase inhibitors (PARPis) against metastatic high-grade serous ovarian carcinoma (HGSOC) is limited to cases with deficient a homologous recombination (HR). Our objective was to determine whether the impairment of the fractalkine receptor (CXCR1) could sensitize HR-proficient cases to PARPis.
Methods: The efficacy of a dual drug combination, including AZD8797, an inhibitor of CXCR1, and several PARPis was examined using cell lines and xenograft models.
Peroxide test strip (PTS) has been widely used for the point-of-care testing (POCT), but its poor sensitivity is a big obstacle for analyzing trace target. Herein, a triple signal amplifying platform integrating the liposome enrichment, the hybridization chain reaction (HCR) circuit, and the image analysis method was constructed to enhance the visual readout of PTS for the sensitive detection of adenosine triphosphate (ATP), an important biomarker of food spoilage. In the presence of ATP, the HCR amplifier was firstly initiated on the surface of magnetic beads (MBs), inducing a large number of glucose oxidase (GOD) imbedded liposomes to be attached.

However, the impact of freeze-thaw cycle events on its storage capacity, particularly on the release of dissolved black carbon (DBC), has remained poorly investigated. This study investigated the release behavior of DBC from biochar pyrolyzed at 300-700 °C during freeze-thaw cycles and their retention capacity in soil.

Embedded or migratory ingested foreign bodies (FBs) may be a disaster when not found intraluminally and can be seriously life-threatening. Endoscopic ultrasound (EUS) has advantages of transmural, close range and real-time imaging. Under its guidance, minimally invasive endoscopic removal of FBs and abscess drainage become available and safe to avoid unnecessary surgery.

Background: Adolescent tennis players encounter critical physical demands, but the lack of comprehensive analysis of training types hampers the selection of optimal training programs. This study aims to conduct a systematic literature review to analyze the effectiveness and limitations of various training types on the physical demands of adolescent tennis players, summarizing the optimal training methods to enhance these physical qualities.
Methods: From March 2024, a comprehensive search was conducted across four electronic databases: SCOPUS, PubMed, EBSCOhost (SPORTDiscus), and Web of Science.
Epstein-Barr virus (EBV) manipulates the ubiquitin-proteasome system and regulators of Bcl-2 family to enable the persistence of the virus and survival of the host cells through the expression of viral proteins in distinct latency patterns. We postulate that the combination of bortezomib (proteasome inhibitor) and venetoclax (Bcl-2 inhibitor) [bort/venetoclax] will cause synergistic killing of post-transplant lymphoproliferative disorder (PTLD) through targeting the pro-survival function of latent viral proteins such as latent membrane protein-1 (LMP-1) and EBV nuclear antigen-3C (EBNA-3C). Bort/venetoclax could synergistically kill spontaneous lymphoblastoid cell lines (sLCLs) derived from patients with PTLD and EBV-associated hemophagocytic lymphohistiocytosis by inducing DNA damage response, apoptosis and G1-S cell cycle arrest in a ROS-dependent manner.

This study investigates the regional distribution, ecological risks, and sources of 44 antibiotics in seawater across four coastal aquaculture areas in Hainan island (Wenchang, Sanya, Danzhou, and Wanning). Among the 44 antibiotics tested across 42 sampling sites, all were detected with a 100 % detection rate.

However, the factors and mechanisms involved in heterochromatin formation and maintenance still remain largely unknown. Here, we show that insulin receptor tyrosine kinase substrate (IRTKS), an I-BAR domain protein, is indispensable for constitutive heterochromatin formation via liquid‒liquid phase separation (LLPS).

This study aimed to explore tPA-related adverse events (AEs) reported in the United States Food and Drug Administration Adverse Event Reporting System (FAERS), assess the potential safety of three preferred tPA therapies for treating AMI, and provide guidance for selecting tPA for prehospital thrombolysis.
Method: Four algorithms, including ROR, PRR, BCPNN, and MGPS, were used to quantify the signals of Tenecteplase, Reteplase, and Alteplase related AEs and compare the differential degrees of the three tPA-associated AEs in the actual data.
Objective: We aimed to investigate the prevalence of vascular complications in acute pancreatitis (AP), to compare patient outcomes using various treatments, and to explore the related risk factors.
Methods: Consecutive AP patients admitted from January 2010 to July 2017 were retrospectively included. Demographics, vascular complications, laboratory indices, and imaging findings were collected.
Background: Pediatric-type diffuse high-grade glioma (pHGG) is the most frequent malignant brain tumor in children and can be subclassified into multiple entities. Fusion genes activating the MET receptor tyrosine kinase often occur in infant-type hemispheric glioma (IHG) but also in other pHGG and are associated with devastating morbidity and mortality.
Methods: To identify new treatment options, we established and characterized two novel orthotopic mouse models harboring distinct MET fusions.
Cadmium (Cd) is a neurotoxic contaminant that induces cognitive decline similar to that observed in Alzheimer's disease (AD). Autophagic flux dysfunction is attributed to the pathogenesis of AD, and this study aimed to investigate the effect of autophagy on environmental Cd-induced AD progression and the underlying mechanism. Here, Cd exposure inhibited autophagosome-lysosome fusion and impaired lysosomal function, leading to defects in autophagic clearance and then to APP accumulation and nerve cell death.

The effect of routine treatment for lesions lightly stained with Lugol's iodine solution is limited, and the addition of natural substances to a regular diet is becoming increasingly common. Dendrobium officinale polysaccharides (DOP), one of the major active components of Dendrobium officinale, have hypoglycemic and hypolipidemic effects and are used clinically to treat diabetes. However, the role of DOP in delaying DKD progression remains unclear.
