The mesenchymal-epithelial transition factor (c-Met) is a tyrosine kinase receptor protein, and excessive cell transformation can lead to cancer. Therefore, there is an urgent need to develop novel receptor tyrosine kinase inhibitors by inhibiting the activity of c-Met protein. In this study, 41 compounds are selected from the reported literature, and the interactions between phenoxy pyridine derivatives and tumor-associated proteins are systematically investigated using a series of computer-assisted drug design (CADD) methods, aiming to predict potential c-Met inhibitors with high activity.

This study focuses on effectively controlling landslides at the boundary of a soft rock open-pit dump while ensuring safe increases in the dump's capacity and optimal utilization of external dump sites. To achieve this, the adoption of a local filling method for the dump base is proposed. We construct a spacecraft performance-fault relationship graph of the control system, which can help space robots locate and repair spacecraft faults quickly. In order to improve the performance-fault relationship graph, we improve the Deep Deterministic Policy Gradient (DDPG) algorithm, and propose a relationship prediction method that combines representation learning reasoning with deep reinforcement learning reasoning. We take the spacecraft performance-fault relationship graph as the agent learning environment and adopt reinforcement learning to realize the optimal interaction between the agent and the environment.

Clinical efficacy of treatments against non-obstructive azoospermia (NOA), which affects 1% of men, are currently limited by the incomplete understanding of NOA pathogenesis and normal spermatogenic microenvironment. Here, we profile >80,000 human testicular single-cell transcriptomes from 10 healthy donors spanning the range from infant to adult and 7 NOA patients. We show that Sertoli cells, which form the scaffold in the testicular microenvironment, are severely damaged in NOA patients and identify the roadmap of Sertoli cell maturation.

In the past few years, Leydig cell (LC) transplantation has been regarded as an effective strategy for providing physiological patterns of testosterone in vivo. Recently, we have successfully converted human foreskin fibroblasts (HFFs) into functional Leydig-like cells (iLCs) in vitro by using the CRISPR/dCas9 system, which shows promising potential for seed cells.
