Sleep deprivation (SD) has been reported to induce intestinal damage by several mechanisms, yet its role in modulating epithelial repair remains unclear. In this study, we find that chronic SD leads to colonic damage through continuous hypoxia. However, HIF1α, which generally responds to hypoxia to modulate barrier integrity, was paradoxically dysregulated in the colon.

  • Abdominal aortic aneurysm (AAA) is a serious condition that predominantly affects older men, but the reasons for its increased prevalence with age are not well understood.
  • Researchers conducted RNA sequencing on mouse aortas from young and old males, alongside human data, to identify gene changes related to age and AAA, discovering 1,001 genes with age-related transcriptional variations.
  • Their findings suggest that aging impacts mitochondrial function and extracellular matrix (ECM) organization, linking these changes to the development of AAA, with aaortic conditions induced by angiotensin II mirroring the ECM alterations seen in older mice.

  • The study investigates the effects of geraniol acetate (gefarnate) in reducing lipid levels in patients with coronary artery disease (CAD) who still experience high triglycerides after statin treatment.
  • Patients in a trial received either gefarnate with statins or just statins, and results showed significant reductions in triglyceride and increases in HDL cholesterol after one month of treatment with gefarnate.
  • These findings suggest that gefarnate might help manage CAD by lowering harmful lipid levels while simultaneously protecting against gastrointestinal injuries caused by aspirin.

Background: The interaction between the intestinal flora and gastroenteropancreatic neuroendocrine neoplasms (GEP-NENs) remains poorly understood, despite the known effect of the gut microbiota on gastrointestinal adenocarcinomas. Hence, the present research aimed to determine the potential causal correlation between the intestinal flora and GEP-NENs by conducting a bidirectional Mendelian randomization (MR) analysis.

Methods: Two-sample MR analysis was conducted using the summary statistics of the gut microbiota from the MiBioGen consortium and those of GEP-NENs from the FinnGen research project.

Minimally processed fruits are increasingly demanded in modern society, but the management of perishable waste pomaces (WPs) and the products' short shelf-life are still big issues. Here, a facile approach of reconstruing apple pomace (AP) into edible bio-nanocomposite coatings of fresh-cutting apple slices was successfully developed through alkaline demethylation followed by high-pressure homogenization. The fibrillation of AP fibers is largely improved by -COO at a concentration of 1.

A cascade Nazarov cyclization/dicycloexpansions reaction was developed for the precise synthesis of the angularly fused M/5/N (M=5, 6; N=4-9, 13) tricyclic skeletons. The prioritized expansion of the first ring played a critical role in the transformations, due to the release of ring strain, and the nature of the substituents present on the substrate is another influencing factor. This pioneering cascade reaction features broad substrates scope (33 examples), short reaction time, exceptional yields (up to 95 %), and remarkable regioselectivities (>20 : 1).

Glioma, a malignant and infiltrative neoplasm of the central nervous system, poses a significant threat due to its high mortality rates. Branched-chain amino acid transaminase 1 (BCAT1), a key enzyme in branched-chain amino acid (BCAA) catabolism, exhibits elevated expression in gliomas and correlates strongly with poor prognosis. Nonetheless, the regulatory mechanisms underlying this increased BCAT1 expression remains incompletely understood.

The oxidation of benzylic alcohols is an important transformation in modern organic synthesis. A plethora of photoredox protocols have been developed to achieve the aerobic oxidation of alcohols into carbonyls. Recently, several groups described that ultraviolet (UV) or purple light can initiate the aerobic oxidation of benzylic alcohols in the absence of an external catalyst, and depicted different mechanisms involving the photoinduction of O as a critical reactive oxygen species (ROS).

An asymmetric intramolecular spiro-amination to high steric hindering -C-H bond of 1,3-dicarbonyl via nitrene transfer using inactive aryl azides has been carried out by developing a novel Cp*Ir(III)-SPDO (spiro-pyrrolidine oxazoline) catalyst, thereby enabling the first successful construction of structurally rigid spiro-quaternary indolinone cores with moderate to high yields and excellent enantioselectivities. DFT computations support the presence of double bridging H-F bonds between [SbF] and both the ligand and substrate, which favors the plane-differentiation of the enol π-bond for nitrenoid attacking. These findings open up numerous opportunities for the development of new asymmetric nitrene transfer systems.

Article Synopsis
  • Sleep deprivation (SD) leads to gut damage primarily through reactive oxygen species (ROS) accumulation, but specific early events impact gut health, especially goblet cells, before ROS levels increase.
  • The study found that mice suffering from SD showed high levels of metabolites related to ferroptosis, a type of cell death, indicating a connection between SD and gut damage.
  • Melatonin was shown to counteract this ferroptotic damage by reducing lipid peroxidation linked to ALOX15 signaling, and ALOX15 knockout also protected mice, suggesting melatonin and ferroptosis as potential targets for preventing gut damage from SD.*
