  • A multidrug-resistant bacterial strain, LS81, was isolated from a clinical patient's abdominal drainage fluid in China and characterized for its resistance genes.
  • LS81 tested positive for specific carbapenemase genes located on a plasmid and was capable of transferring these genes to other strains.
  • Whole-genome sequencing identified LS81 as belonging to sequence type 196 (ST196) and revealed a complex accessory genome, highlighting the importance of ongoing surveillance for this emerging hospital-acquired pathogen.

Article Synopsis
  • Jiaotou (Allium chinense) is a popular herb from China known for its culinary and medicinal uses, and recently, a significant disease resembling anthracnose has been detected in Hunan Province affecting its growth.
  • The symptoms of this disease include yellow to brown spots on the leaves, leading to distortion and wilting, with a high incidence rate of around 50% over a 20-hectare area.
  • Pathogen isolation from affected plants revealed fungal characteristics similar to Colletotrichum spaethianum, confirmed by genetic sequencing of the internal transcribed spacer (ITS) and several other genes.

Imbalanced Sirtuin 1 (SIRT1) levels may lead to liver diseases through abnormal regulation of autophagy, but the roles of SIRT1-regulated autophagy in hepatocellular carcinoma are still controversial. In this study, we found that SIRT1 mRNA and protein levels were upregulated in hepatocellular carcinoma, and high SIRT1 expression hinted an advanced stage and a poor prognosis. The differentially expressed proteins were significantly elevated in autophagy, cellular response to stress, and immune signaling pathways.

Colletotrichum fructicola is a globally significant phytopathogenic fungus. Mycovirus-induced hypovirulence has great potential for biological control and study of fungal pathogenic mechanisms. We previously reported that the mycovirus Colletotrichum alienum partitivirus 1 (CaPV1) is associated with the hypovirulence of C.

Saline soils are widely distributed in arid areas but there is a lack of mechanistic understanding on the effect of salinity on the formation and biochemical composition of soil organic carbon (SOC). We investigated the effects of salinity on the accumulation of microbial necromass under natural vegetation and in cropland in salt-affected arid areas stretching over a 1200-km transect in northwest China. Under both natural vegetation and cropland, microbial physiological activity (indicated by microbial biomass carbon normalized enzymatic activity) decreased sharply where the electrical conductivity approached 4 ds m (a threshold to distinguish between saline and non-saline soils), but microbial biomass was only slightly affected by salinity.

Objective: This study aimed to investigate the impact of microbubble degradation and flow velocity on Sub-Harmonic Aided Pressure Estimation (SHAPE), and to explore the correlation between subharmonic amplitude and pressure as a single factor.

Methods: We develop an open-loop vascular phantom platform system and utilize a commercial ultrasound machine and microbubbles for subharmonic imaging. Subharmonic amplitude was measured continuously at constant pressure and flow velocity to assess the impact of microbubble degradation.

It is significant to understand the adsorption mechanisms of shale gas (CH) and CO in shale formations to enhance CH recovery rates and enable geological CO storage. This study provides a comprehensive investigation into the adsorption behaviors of CO and CH within dry and hydrous calcite nanopores, utilizing a combination of grand canonical Monte Carlo simulations, molecular dynamics simulations, and density functional theory calculations. In dry calcite slits, the calculated results for the adsorption capacity, density profile, and isosteric heat of CO and CH reveal that CO possesses a stronger adsorption affinity, making it preferentially adsorb on the pore surface compared to CH.

Rhizoctonia solani is a widespread and devastating plant pathogenic fungus that infects many important crops. This pathogen causes tobacco target spot, a disease that is widespread in many tobacco-growing countries and is destructive to tobacco. To identify antagonistic microorganisms with biocontrol potential against this disease, we isolated Streptomyces strains from forest inter-root soil and screened a promising biocontrol strain, ZZ-21.

This study aims to evaluate the tolerance of endophytic fungi isolated from the fibrous roots of Polygonatum kingianum to arsenic (As) and cadmium (Cd) and their physiological response mechanisms. Five isolated strains were obtained with EC values for As(V) ranging from 421 to 1281 mg/L, while the other three strains tolerated Cd(II) with an EC range of 407-1112 mg/L. Morphological and molecular identification indicated that these eight strains were Cladosporium spp.

Article Synopsis
  • The tumor microenvironment is complex and includes tumor-associated macrophages (TAMs), which can be influenced by tumor cells to create an environment that supports tumor growth.
  • This study found that liver cancer (HCC) cells cause macrophages to change into a M2-like phenotype, which is associated with promoting cancer, and that this change is linked to the role of autophagy in macrophage behavior.
  • Inhibiting autophagy in macrophages leads to M2 polarization through a specific mechanism involving the NF-κB pathway, highlighting a potential target for cancer treatment by disrupting this polarization process.

Millions of adenosine (A) to inosine (I) RNA editing events are reported and well-studied in eukaryotes; however, many features and functions remain unclear in prokaryotes. By combining PacBio Sequel, Illumina whole-genome sequencing, and RNA Sequencing data of two Klebsiella pneumoniae strains with different virulence, a total of 13 RNA editing events are identified. The RNA editing event of badR is focused, which shows a significant difference in editing levels in the two K.

Article Synopsis
  • The study investigates the role of autophagy in macrophages within the tumor microenvironment of hepatocellular carcinoma (HCC), finding reduced autophagy linked to worse patient outcomes and increased metastasis.
  • Researchers identified that HCC triggers decreased autophagy in macrophages by activating mTOR, which in turn promotes cancer progression through mechanisms involving the NLRP3 inflammasome and IL-1β release.
  • Targeting the signaling pathways related to IL-1β showed potential as a therapeutic strategy, indicating that disrupting the feedback loop between autophagy inhibition and macrophage recruitment could help treat HCC more effectively.

To assess the performance of biplane area-length method in measuring left atrial (LA) volume and sphericity index and to investigate the correlation of LA reservoir function and sphericity index with atrial fibrosis on cardiac magnetic resonance imaging (MRI) in patients with mitral valve disease (MVD). Forty-eight patients with MVD undergoing cardiac MRI scan were enrolled in this retrospective study. LA reservoir function, measured as maximum volume (LAV), minimum volume (LAV) and ejection fraction (LAEF), and sphericity index were quantified using the biplane area-length method and standard short-axis approach, respectively.

The Tibetan Plateau (TP) is a sensitive alpine environment of global importance, being Asia's water tower, featuring vast ice masses and comprising the world's largest alpine grasslands. Intensified land-use and pronounced global climate change have put pressure on the environment of the TP. We studied the tempo-spatial variability of dissolved organic matter (DOM) to better understand the fluxes of nutrients and energy from terrestrial to aquatic ecosystems in the TP.

Background: While the correlation between PD-L1 expression and KRAS mutation has been previously reported in other solid tumors such as non-small cell lung cancer (NSCLC), whether PD-L1 can be modulated by ERK signaling downstream of KRAS in intrahepatic cholangiocarcinoma (iCCA) and the underlying molecular regulatory mechanism remain unclear.

Methods: The expression of ERK, p-ERK, PD-L1 and autophagy markers following KRAS knockdown or Ras/Raf/MEK/ERK signaling inhibitors treatment was examined in two human iCCA cell lines (HuCCT1 and RBE) using western blotting and immunofluorescence. Both pharmacological autophagy inhibitors and short-interfering RNA against ATG7 were applied to inhibit autophagy.
