Fluorescent in situ hybridization has been the definitive modality in testing for overexpression of the Human Epidermal Growth Factor Receptor 2 (HER2) for decades to guide the appropriate treatment for cancer patients. In more recent years innovation and new techniques have been developed to supplant or even replace FISH as a standard method for biomarker testing. Alternative testing methods such polymerase chain reaction (PCR), next-generation sequencing (NGS), and other in situ hybridization (ISH)-derived techniques such as chromogenic-ISH (CISH) have been shown in multiple publications to have high concordance with FISH in addition to advantages in economics, logistics and practicality to the point where CISH and derived methods appear to have eclipsed FISH as a testing method of choice after immunohistochemistry (IHC).

The purpose of the current research effort was two-fold: (a) determine what behaviors successful veteran job candidates perform during the job application process that distinguish them from less successful ones, and (b) identify the behaviors of successfully employed veterans that distinguish them from those that are less successful or unable to maintain civilian employment. To address these issues, a research initiative was undertaken by the City of Clarksville, TN; Montgomery County, TN; the Tennessee Department of Labor; and the Tennessee Department of Veterans Services. Researchers conducted a qualitative study in the environs of Nashville, TN and Clarksville, TN, where Fort Campbell a United States Army installation, is located.

The primary goal of this investigation is to determine the feasibility of using vibrational spectroscopy techniques for the analysis of soil samples in order to characterize a specific area and hence create a soil map for use in forensic investigation. We compare spectra of "artifacts" which are exclusive to a single location or area rather than focus on the general intrinsic components of soil. Comparison of these artifacts may allow the creation of a map of different regions and thus allow matching of soil evidence to these regions (spectra).

Objective: Sex trafficking is an unrelenting problem in Mississippi. No quantitative data currently exist on the prevalence of sex trafficking or the identification of victims in the state.

Method: This study used the Trafficking in Victims Identification Tool (TVIT) (Short Version) to identify the extent to which a sample of clients (n = 28) receiving services at a non-profit social services agency in Jackson, Mississippi, were also victims of sex trafficking.

Identifying potential modifiable determinants of functional movement ability and fatigue may inform efforts to maintain constant physical readiness, especially in active duty military over 40 years of age, who are largely sedentary throughout their work day. Mitotic progression in eukaryotic cells depends upon the activation of cyclin-dependent kinase 1 (CDK1), followed by its inactivation through the anaphase-promoting complex (APC)/cyclosome-mediated degradation of M-phase cyclins. Previous work revealed that expression of a constitutively active CDK1 (CDK1AF) in HeLa cells permitted their division, but yielded G1 daughter cells that underwent premature S-phase and early mitotic events. While CDK1AF was found to impede the sustained activity of APC-Cdh1, it was unknown if this defect improperly stabilized mitotic substrates and contributed to the occurrence of these premature M phases.

Recent decades have witnessed an increase in biological explanations of common disorders such as depression and anxiety, a phenonmenon termed "medicalization" (Conrad, 2007). Behavior analysts may find it difficult to implement non-drug treatment with a populace that has become inundated with biological explanations and a preference for medication as the treatment of choice. Steers and heifers were generated from Angus (A) and Belgian Blue (BB) sires mated to Brahman x Hereford (B x H) F(1) cows to characterize their growth, carcass traits, and LM shear force. A total of 120 B x H cows purchased from 2 herds and 35 bulls (14 A and 21 BB) produced calves during the 5-yr project. After the stocker phase, a representative sample of A- and BB-sired heifers and steers were transported to the Iberia Research Station to be fed a high-concentrate diet.

The influence of horn fly control with commercially available ear tags was studied on beef replacement heifers (n = 670) for growth and reproductive performance. The study was conducted at five sites in Louisiana over 3 yr. Heifers used were yearling replacement females that were exposed to fertile bulls during a limited spring breeding season that coincided with the horn fly season.

A study was conducted to compare Brangus, Beefmaster, Gelbray, and Simbrah breed influences for economically important traits. Brangus (9), Beefmaster (12), Gelbray (10), and Simbrah (7) sires were used in purebred and crossbred (Brahman x Hereford F1 cows) matings to generate calves (326) in eight breed groups. Beefmaster cows were of similar size (448 kg), Brangus and Gelbray cows were 11% heavier (501 and 503 kg), and Simbrah cows were 21% heavier (548 kg) compared to Brahman x Hereford F1 cows (452 kg).

In a prospective study of 42 patients with myelofibrosis with myeloid metaplasia (MMM), peripheral blood (PB) and bone marrow (BM) interphase cytogenetics and PB CD34 enumeration were performed concomitantly with BM karyotype analysis. Interphase cytogenetics was performed with a panel of fluorescence in situ hybridization (FISH) probes that were capable of detecting most of the known recurrent cytogenetic lesions in MMM. There was a close concordance in the results of interphase cytogenetics between PB and BM, regardless of the PB CD34 count.

Mantle-cell lymphoma (MCL) has a poorer prognosis than other small B-cell lymphomas, thus a definitive diagnosis is essential. The t(11;14)(q13;q32) associated with MCL juxtaposes portions of CCND1 (11q13) and IGH (14q32), resulting in over-expression of cyclin D1. In this study, a highly sensitive two-colour fluorescence in situ hybridization (FISH) method was developed to detect t(11;14)(q13;q32) in nuclei isolated from paraffin-embedded tissue.

The goal of this work was to optimize dendritic cell (DC) preparations obtained from patients suffering from chronic myeloid leukemia (CML) and compare them with DC prepared from normal CD14+ mononuclear cells (MNC). We studied normal DC and bcr-abl+ leukemic DC (CML-DC) yields, expression of membrane molecules, differentiation status, and ability to stimulate T cells. We isolated DC precursors from PBMC by CD14-specific immunoadsorption and cultured them for 7 days in GM-CSF and IL-4, followed by a 3-day incubation to fully differentiate the cells.

In the bovine oviduct, monosaccharides may play a role in the preparation of gametes for fertilization. Sperm are sequestered in the isthmic region of the oviduct where capacitation, requisite biochemical changes in sperm membranes, may take place. Retention of spermatozoa in the oviductal isthmus is dependent on a carbohydrate recognition system between oviductal epithelium and sperm membrane lectins.

Buccal smear analysis is a noninvasive, fast, and relatively inexpensive diagnostic method. It is used commonly where rapid gender identification is necessary or, more recently, for detection of aneusomy, microdeletion syndromes, and a variety of polymerase chain reaction-based molecular genetic tests.
