A method is presented for the quantitative analysis of early insulin release in man. There were measured arterial insulin levels after glibornuride administered intravenously. The mathematical procedure has been modified: Modification I is based on the assumption that early insulin release represents a wave like insulin delivery, modification II is based on the assumption that this insulin bolus is immediately followed by a slower insulin release which must be distinguished from the second phase of insulin release.
View Article and Find Full Text PDF22 outpatients with primary hyperlipoproteinemia type IIb and IV were treated in periods of eight weeks as follows: placebo; 0,15 g phenformin/day; 0,15 g phenformin + 1,5 g clofibrate/day; 1,5 g clofibrate/day; placebo. Compared to the first placeboperiode the serumtriglycerides were significantly lowered by phenformin (about 26%), by the combined treatment with phenformin + clofibrate (60%) and by clofibrate (51%) after eight weeks of treatment. The serumcholesterol was significantly lowered by phenformin (10%) and by the combined treatment with phenformin and clofibrate (14%), but not significantly by clofibrate (8%).

Two different commercial preparations of clofibrate and one preparation of beta-pyridylcarbinol were each given for two months separated by placebo periods of two weeks. All three drugs were equally effective against increased cholesterol levels.

23 patients with hyperlipidemia and hyperuricemia received acetamidoethyl-(4-chlorophenyl)-(trifluoromethylphenoxy)-acetate (halofenate), a clofibrate derivative, and probenecid or probenecid and placebo over 36 weeks following a placebo period of 6 weeks. Halofenate compared with probenecid lowered elevated serum uric acid levels satisfactorily to a therapeutic level between 5 and 6 mg/100 ml. Serum triglyceride levels were not always lowered sufficiently, serum cholesterol levels were not influenced.

The increase in serum triglyceride levels in healthy subjects after a diet rich in carbohydrates in influenced by several factors such as energy balance, absolute quantity of carbohydrate, individual response of the test person, etc. With the aid of a dietary formula trial in healthy adults, saccharose consumed in usual quantities is shown to cause no increase in triglyceride or cholesterol levels.
View Article and Find Full Text PDFLipoatrophic diabetes is characterized by: complete lack of subcutaneous fattissue (generalized lipodystrophy), insulin resistant diabetes mellitus, hepatosplenomegaly, excessive hyperthyroidism, elevated basal metabolic note without hyperthyroidism. Recently, "cystic" alterations in the bone have been described as a possible further characteristic. The case of a young woman, now aged 21, who has had generalized lipodystrophy for 17 years and overt diabetes mellitus for 6 years is reported.
