Introduction: Vestibular schwannomas are benign tumors that account for 8% of all intracranial tumors. Due to their location in the cerebellopontine angle and internal auditory canal, access to these lesions requires disruption of the temporal bone. The pneumatization of this bone varies between individuals, and literature reports suggest that it may influence the frequency of cerebrospinal fluid leaks.

Unruptured giant intracranial aneurysms (GIA) are those with diameters of 25 mm or greater. As aneurysm size is correlated with rupture risk, GIA natural history is poor. Parent artery occlusion or trapping plus bypass revascularization should be considered to encourage intra-aneurysmal thrombosis when other treatment options are contraindicated.

Herein, we describe a two-step sequential flow synthesis: Pd-catalyzed aerobic oxidation to an aldehyde 2, which is then converted by reductive amination in H-Cube® PRO into CPL302415 (3). CPL302415 is our new PI3Kδ inhibitor, which is now under evaluation for the treatment of systemic lupus erythematosus. The process was optimized using the DoE approach and generalized to other biologically active derivatives of CPL302415.

Giant intracerebral aneurysms (GIA) comprise up to 5 % of all intracranial aneurysms. The indirect surgical strategy, which leaves the GIA untouched but reverses the blood flow by performing a bypass in combination with proximal parent artery occlusion is a useful method to achieve spontaneous aneurysm occlusion. The goal of this study was to assess the utility of computational fluid dynamics (CFD) in preoperative GIA treatment planning.

Biological invasions cause biodiversity erosion on a global scale. Invasive species spreading beyond their natural range compete with native fauna for food and space, push native species to suboptimal habitats, impairing their behaviour and thus limiting their occurrence. Freshwater ecosystems are especially vulnerable to biological invasions and their ecological and economic impacts.

  • - BTK inhibitors have become crucial in treating chronic lymphocytic leukemia (CLL) by disrupting B-cell signaling, which inhibits the growth and survival of cancer cells.
  • - These inhibitors, particularly Ibrutinib and newer options like acalabrutinib and zanubrutinib, are effective for both new patients and those with advanced or difficult-to-treat CLL, especially high-risk cases.
  • - However, continuous use of BTK inhibitors can lead to resistance, with certain genetic mutations identified as key factors in this resistance, impacting treatment outcomes.

  • Extended aortic repair is essential for maintaining long-term surgical success in young patients with DeBakey type 1 aortic dissection, as they face higher risks of aortic degeneration due to their longer life expectancy.
  • A study analyzed 1,199 patients under 60 who had aortic repair surgeries across Europe from 2005 to 2021, comparing outcomes between ascending aortic repair and total aortic arch repair techniques.
  • Results showed no significant difference in the need for additional surgeries after 5 years between the two techniques, but total aortic arch repair had higher rates of postoperative complications like stroke and dialysis, while overall mortality rates were similar.

Delayed cerebral ischemia (DCI) is a serious, life-threatening, complication affecting patients who have survived the initial bleeding from a ruptured intracranial aneurysm. Due to the challenging diagnosis, potential DCI prognostic markers should be of value in clinical practice. According to recent reports isoprostanes and red blood cell distribution (RDW) showed to be promising in this respect.

  • The study investigates the effects of different arterial cannulation methods (femoral, supra-aortic, and direct aortic) on outcomes for patients undergoing surgery for acute Stanford type A aortic dissection (TAAD).
  • Results show that in-hospital mortality rates for patients using femoral or supra-aortic cannulation are similar, while direct aortic cannulation yields lower mortality rates compared to femoral cannulation.
  • Additionally, switching cannulation sites during surgery increases the risk of in-hospital mortality, while long-term outcomes (10-year mortality) remain consistent across the groups.

Recent events in conflict zones have emphasized that the successful detection and characterisation of buried clandestine complexes, bunkers and tunnels is vitally important for forensic investigators globally, to reduce or solve criminal activities, address national security threats and avoid potential terrorist attacks. However, this can often prove very difficult, particularly in urban areas, with potentially both below-ground non target items and above-ground infrastructures present, that can interfere with detecting target(s). Here we provide selected successful case studies where forensic geoscience techniques were used to detect and characterise buried clandestine complexes, bunkers and tunnels using different geophysical techniques.

Glioblastoma, a highly aggressive brain tumor, poses significant treatment challenges. A deeper investigation into its molecular complexity is essential for the identification of novel prognostic biomarkers and therapeutic strategies, potentially improving patient outcomes in terms of survival and quality of life. While nuclear DNA mutations have been extensively studied, the role of mitochondrial DNA (mtDNA) mutations, specifically in the D-loop region, remains poorly understood.

Article Synopsis
  • Surgery for type A aortic dissection (TAAD) has a high early mortality risk, with in-hospital mortality rates increasing based on the urgency of the procedure, ranging from 10% for urgent surgeries to over 57% for the most critical salvage procedures.
  • A multicenter study using data from the European Registry of TAAD (ERTAAD) found that preoperative arterial lactate levels correlate with procedure urgency, indicating that more critical cases are associated with poorer outcomes.
  • The urgency classification enhances the ability to predict patient risk, suggesting that despite high mortality in severe cases, salvage surgeries are often justified as many patients can still survive to discharge.
