Analysis of beta, gamma, and delta T-cell receptor genes in mycosis fungoides and Sezary syndrome.

The authors have analyzed the configuration of immunoglobulin (Ig) and beta, gamma and delta T-cell receptor (TCR) genes in DNA extracted from skin, lymph nodes, and peripheral blood mononuclear cells obtained from 41 patients with mycosis fungoides (MF), 14 patients with Sezary syndrome, and 13 patients with benign inflammatory dermatoses. No discrete rearranged bands (DRB) were detected in patients with inflammatory dermatoses. In tissue DNA from 19 patients with MF DRB were detected with beta and gamma, but not delta TCR probes.

Isolation and ultrastructural analysis of microfibrillar structures from foetal bovine elastic tissues. Relative abundance and supramolecular architecture of type VI collagen assemblies and fibrillin.

Extensive intact assemblies of matrix macromolecules have been solubilized from foetal calf skin, nuchal ligament and aorta by a new procedure that includes bacterial collagenase digestion under non-reducing, non-denaturing conditions and gel filtration chromatography. Type VI collagen was identified as the major microfibrillar element of these tissues by SDS-PAGE analysis and Western blotting. Rotary shadowing electron microscopy of these preparations revealed by far the most abundant and extensive arrays of intact collagen VI microfibrils isolated to date.

Effects of albendazole and albendazole sulfoxide on cultures of differentiating rodent embryonic cells.

Micromass cell culture systems for rat embryo midbrain (CNS) and limb bud (LB) cells were employed to assess the in vitro developmental toxicity of the human and veterinary anthelmintic albendazole (ABZ) and its sulfoxide metabolite (SOABZ). ABZ is reported to be teratogenic in rats, and is extensively metabolized to the sulfoxide derivative. It has been postulated that SOABZ is the reactive metabolite responsible for albendazole's developmental toxicity and anthelmintic activity in vivo.

Analysis of beta, gamma, and delta T-cell receptor genes in lymphomatoid papulosis: cellular basis of two distinct histologic subsets.

To investigate the histogenesis of lymphomatoid papulosis (LYP), we have analyzed the configuration of the beta, gamma, and delta T-cell receptor (TCR) and Ig genes in DNA from 31 biopsies of 18 patients with this condition and also from peripheral blood mononuclear cells of eight of these patients. Immunoglobulin genes were in a germ-line configuration in all patients, and TCR genes were in a germ-line configuration in six patients. In nine patients, one or two rearranged bands (RB) were detected with both beta and gamma TCR probes and in one patient with beta, gamma, and delta TCR probes.

Prior research has suggested that two types of fast eye movements (FEMs) can be distinguished behaviorally. Foveating saccades respond to salient peripheral targets by directing the target image to the fovea. Cytochemical localization of mercury in Saccharomyces cerevisiae treated with mercuric chloride.

We describe a cytochemical method for localizing mercury at the electron microscopic level in the yeast Saccharomyces cerevisiae. After addition of a lethal concentration of mercuric chloride to growing yeast cells, mercury was associated with the cell wall and cytoplasmic membrane. Little or no mercury was present in the cytoplasm.

Pineal and CNS germ cell tumors: Royal Marsden Hospital experience 1962-1987.

A retrospective analysis has been made of all patients with pineal and CNS germ cell tumors who were treated at The Royal Marsden Hospital between 1962-1987. A total of 67 new cases were seen: 17 had initial histological verification of tumor type and the remainder were tested for radiosensitivity with a dose of 20 Gy following a shunting procedure. Patients with germ cell or radiosensitive tumors were treated with a uniform policy of whole neuraxis radiotherapy giving 50 Gy to the local tumor and 30 Gy to the remaining brain and spinal cord.

Plasma exchange in refractory cutaneous vasculitis.

Eight patients with intractable cutaneous leucocytoclastic vasculitis were given a trial of plasma-exchange therapy. All but one improved, five substantially. Four have been successfully treated by intermittent plasma exchange for periods of 5-12 years.

Foveating saccades.

A review of the literature revealed that foveating saccades were found to be faster than other fast eye movements (FEMs) except optokinetic nystagmus (OKN) quick phase. In the present experiment, foveating saccades were compared to OKN quick phase in humans and were found to have higher maximum speeds and shorter durations. Unlike previous experiments, foveating saccades were made to targets at unpredictable distances, and active pursuit during OKN was discouraged.

Statistical methods are considered for analysis of data arising from a mitotic chromosome loss assay in Saccharomyces cerevisiae strain D61.M. The methods make use of reproducibility trial data from the assay (presented herein) and previous data, which suggest a unimodal, 'umbrella-patterned' dose response.

Two patients who were concomitantly undergoing methadone maintenance treatment and receiving phenytoin suffered continuing opioid withdrawal symptoms as a results of phenytoin's acceleration of methadone metabolism. When anticonvulsant therapy was discontinued, these patients had seizures. One patient was switched to carbamazepine as an alternative anticonvulsant, but this treatment resulted in a similar clinical problem.

Solitary morphoea profunda.

This study describes a series of patients with an unusual form of scleroderma. All five patients presented clinically with solitary, indurated plaques involving the upper trunk. Histologically the lesions were characterized by a dense, mononuclear cell infiltrate mainly in the subcutis, which was rich in plasma cells, and was associated with marked sclerosis and hyalinization of the connective tissue at this level.
