Isolation and characterization of thymidylate synthetase mutants of Xanthomonas maltophilia.

Thymidylate synthetase mutants of Xanthomonas maltophilia ATCC 13270 were isolated on a solid minimal medium containing 50 mg/l thymidine and a high concentration of trimethoprim (500 mg/l). It was found that a high concentration of trimethoprim was required to prevent background growth of the wild-type strain. The isolated mutants could grow on thymidine or dTMP at a concentration of 50 mg/l while they were unable to grow on 1000 mg/l thymine or 50 mg/l deoxyuridine.

Clinical safety and efficacy of once-a-day amlodipine for chronic stable angina pectoris.

This study evaluated the safety, antianginal and antiischemic effects of amlodipine, a dihydropyridine calcium antagonist, in patients with chronic stable angina pectoris. The patients (n = 29) were evaluated during a 26-week single-blind dose titration phase followed by a 6-week double-blind placebo-randomized withdrawal phase. No patient withdrawals resulted from adverse events directly related to amlodipine.

Metabolism of pyrimidine bases and nucleosides by Pseudomonas fluorescens biotype F.

Pyrimidine metabolism in Pseudomonas fluorescens biotype F, and its ability to grow in liquid culture on pyrimidines and related compounds was investigated. It was found that uracil, uridine, cytosine, cytidine, deoxycytidine, dihydrouracil, dihydrothymine, beta-alanine or beta-aminoisobutyric acid could be utilized by this pseudomonad as a sole nitrogen source. Only uridine, cytidine, beta-alanine, beta-aminoisobutyric acid or ribose were capable of supporting its growth as a sole source of carbon.

Evaluation of a Cordia-IC enzyme-linked immunosorbent assay kit for the detection of circulating immune complexes.

A commercial kit (Cordia-IC) from Cordis Laboratory, Miami, Fla., was compared with the Raji cell radioimmunoassay for its ability to detect circulating immune complexes (CIC) in sera from 30 control subjects and 118 patients with infectious diseases. The 118 patients were categorized into the following groups: (i) 23 patients with bacterial endocarditis, (ii) 41 patients with bacteremia from an infected intravascular catheter or access device, and (iii) 54 patients with Staphylococcus aureus bacteremia related to a deep tissue infection.

Adenovirus type 2 encephalitis and concurrent Epstein-Barr virus infection in an adult man.

Adenovirus type 2 was isolated from brain tissue obtained by a biopsy and there was a concurrent fourfold rise in the antibody titer. In addition, an initial negative result from a slide test for infectious mononucleosis heterophil antibodies (Monospot) test converted to positive and there was an increase in the IgG antibody titer to Epstein-Barr virus (EBV) viral capsid antigen.

Enzyme-linked immunosorbent assay for detection of immunoglobulin G and M antibodies to teichoic acid in intravascular staphylococcal disease.

An enzyme-linked immunosorbent assay (ELISA) for detection of IgG and IgM antibodies to cell-wall teichoic acids of Staphylococcus aureus and three defined coagulase-negative staphylococci was tested using serum samples from 11 cases of intravascular coagulase-negative staphylococcal infections, 13 cases of Staphylococcus aureus endocarditis, and 24 patients with no evidence of infection. IgG antibody titers to all four teichoic acids in the 13 patients with Staphylococcus aureus endocarditis were significantly different from those in noninfected control patients (p less than 0.0001).

Freeze-dried bone and coralline implants compared in the dog.

Partially demineralized, antigen-extracted, freeze-dried cadaver bone and a hydroxyapatite replica of a marine coral skeleton were implanted in 30 surgically created two-wall intrabony pockets in two dogs. Results after 6 and 8 months showed connective tissue encapsulation and some periodontal ligament formation around the cadaver bone. In contrast, the coralline implant was consistently invaded by fibrovascular tissue, showed bone formation on its internal surfaces and had some periodontal ligament formation around it.

A Salmonella typhimurium strain defective in uracil catabolism and beta-alanine synthesis.

A selection procedure for uracil catabolism mutant strains involving indicator dye plates was developed. Using this method, a strain defective in uracil catabolism has been isolated in Salmonella typhimurium that was temperature-sensitive at 42 degrees C where it required low concentrations of N-carbamoyl-beta-alanine, beta-alanine or pantothenic acid for growth. An extract of the mutant strain degraded uracil at 37 degrees C at a significantly diminished rate compared to that observed for the wild-type strain under the same growth conditions.

Chromatography of aspartate transcarbamoylase from Escherichia coli on agarose-immobilized dyes and alkyl-agaroses of differing carbon length were investigated. The bacterial aspartate transcarbamoylase was bound by Procoin red HE3B-agarose and Cibacron blue F3GA-agarose nearly completely under the conditions chosen relative to other agarose-coupled dyes. The aspartate transcarbamoylase holoenzyme was eluted from the Procion red HE3B-agarose slightly later than from the Cibacron blue F3GA-agarose during salt gradient elution.
