Cell-permeable ubiquitin (Ub) probes labeled with a fluorescent or biotin tag have become effective tools for studying the activity of deubiquitinating enzymes (DUBs) in living cells. However, such probes are usually obtained through total chemical synthesis. In this study, we develop a Sortase A (SrtA)-mediated semi-synthetic strategy for preparing cell-permeable Ub-propargylamide (Ub-PA) with a fluorescent or biotin tag.

Spinal cord organoids (SCOs) are in vitro models that faithfully recapitulate the basic tissue architecture and cell types of the spinal cord and play a crucial role in developmental studies, disease modeling, and drug screening. Physiological cues are required for proliferation and differentiation during SCO culture. However, commonly used basement membrane matrix products, such as Matrigel®, lack tissue-specific biophysical signals.

Spatially resolved transcriptomics (SRT) simultaneously measure spatial location, histology images, and transcriptional profiles of cells or regions in undissociated tissues. Integrative analysis of multi-modal SRT data holds immense potential for understanding biological mechanisms. Here, we present a flexible multi-modal contrastive learning for the integration of SRT data (MuCST), which joins denoising, heterogeneity elimination, and compatible feature learning.

Our study elucidates the role of FOXP1 in chemoresistance in small cell lung cancer(SCLC). FOXP1 enhances chemoresistance by regulating SP8 expression through its super-enhancer (SP8-SE), with SP8 mediating resistance via the homologous recombination repair (HRR) pathway. We also discovered that FOXP1 forms punctate nuclear structures indicative of liquid-liquid phase separation, crucial for its transcriptional regulation.

Background: The current AJCC TNM staging system for medullary thyroid cancer (MTC) is largely adapted from criteria for differentiated thyroid cancer, which may not fully capture MTC-specific prognostic factors. This study aimed to evaluate the prognostic significance of upper mediastinal lymph node metastasis (LNM) and propose corresponding modifications to the N category and staging system to improve their applicability for MTC.

Methods: We conducted a population-based, retrospective study enrolling patients with MTC from 19 Chinese referral centers.

Biodegradable microplastics (MPs) are proposed as sustainable alternatives to conventional MPs, yet their distinct effects on soil microbial communities and ecological functions remain insufficiently understood. This study compares the impacts of biodegradable polylactic acid (PLA) and conventional polyvinyl chloride (PVC) MPs on soil microbial assemblages and nitrogen cycling. Fluorescein diacetate hydrolase (FDAse) activity was temporarily stimulated by 2 % (w/w) PLA and PVC MPs, while 7 % (w/w) PVC MPs initially inhibited FDAse activity before promoting it.

5-Methylcytosine (5mC) functions as a well-characterized epigenetic DNA mark in nuclear DNA, but its presence in mitochondrial DNA (mtDNA) remains elusive. Here, we report a new and rigorous nuclear DNA (nDNA)-excluding mass spectrometry assay enabling the reliable and accurate identification of 5mC in mtDNA for the first time. First, circular mtDNA is enriched over 809-946-fold by combining alkaline lysis and linear DNA-specific RecBCD cutting; nDNA accounts for ∼12-19% of the DNA remaining after this step.

Background: A20, also known as TNF-α-induced protein 3 (TNFAIP3), is a crucial negative regulator of inflammation and immune responses. Emerging evidence suggests that A20 is involved in the regulation of glucose metabolism and plays a significant role in bone metabolic diseases by inhibiting nuclear factor (NF)-κB activation. However, the potential relationship between serum A20 level and bone mineral density (BMD) in patients with type 2 diabetes mellitus (T2DM) has not been explored.

Background And Aims: The diagnostic value of primary biliary cholangitis (PBC)-specific antibodies in patients with elevated alkaline phosphatase (ALP) and gamma-glutamyl transferase (GGT) levels, and other identifiable causes, was unclear. Our study aimed to determine whether etiological treatments in PBC-specific antibody-positive patients could improve liver biochemical tests, thereby distinguishing them from individuals with PBC.

Methods: We enrolled patients who were positive for PBC-specific antibodies and elevated ALP and/or GGT levels but with other identifiable etiologies.

This study investigated the effects of microwave (MW) reheating, water boiling (WB) reheating, and steaming (ST) reheating on the quality attributes (including reheating loss, moisture content, centrifugal loss, water distribution, color, texture, microstructure, flavor, and taste) of pre-cooked braised chicken (PBC), using a non-reheated group as a control (C). The results showed that the ST group demonstrated the lowest reheating loss, and that ST reheating had the least influence on textural characteristics among all the reheating methods. In addition, the results of the scanning electron microscope (SEM) showed that the integrity of the muscle fibers in the ST group was most comparable to the C group.
