Legumes, a dietary staple for centuries, have seen an influx of conventional and unconventional varieties to cater to human care conscious consumers. These legumes often undergo pretreatments like baking, soaking, or boiling to mitigate the presence of non-proteinogenic amino acids (NPAAs) and reduce associated health risks. The recent tara flour health scare, linked to the NPAA baikiain, emphasizes the need for robust analytical methods to ensure the safety and quality of both traditional and novel plant-based protein alternatives.

Background: The use of illicit drugs and stimulants is a burgeoning socioeconomic problem, ultimately leading to an increase in street crimes and deteriorating human health. The persistent presence of CNS drugs in wastewater can also lead to downstream adverse effects on aquatic wildlife and humans.

Objectives: In the present study, a method was developed for the solid-phase extraction and quantitative liquid chromatography coupled with tandem quadrupole/time-of-flight mass spectrometry (LC-QToF-MS) analysis of CNS stimulant drugs and their metabolites in municipal wastewater.

Twenty-two compounds, predominantly catechin, taxifolin, and proanthocyanidin derivatives, were isolated and characterized from the methanol extract of Entada elephantina tuberous rhizomes. Five taxifolin-based metabolites, elephantinosides A-E (1-5) and one proanthocyanidin, elephantinoside F (19), were previously undescribed. Their structures were elucidated primarily based on 1D- and 2D-NMR and HRESIMS data.

5-(3',4'-Dihydroxyphenyl)-γ-valerolactone (VL) is a bioactive metabolite resulting from the gut microbial metabolism of proanthocyanidins and flavonoids, known for its health-promoting effects, including antidiabetic and anti-inflammatory activities. Although VL has been observed in different in vivo studies, its pre-absorption, distribution, metabolism, excretion, toxicity (ADMET) properties have rarely been investigated. This study aims to address this gap by evaluating the pre-ADMET properties of VL for the first time.

Objective: To investigate the clinical characteristics of patients with hemophagocytic lymphohistiocytosis (HLH) and quantify the diagnostic value of various indexes in patients with elevated soluble interleukin-2 receptor (sCD25), so as to construct a diagnostic prediction model of HLH.

Methods: The clinical characteristics of 121 patients with elevated sCD25 (≥2 400 U/ml) in the Third Affiliated Hospital of Sun Yat-Sen University were analyzed retrospectively. The patients were divided into HLH group and non-HLH group according to the diagnostic criteria of HLH.

Background: In this study, we aimed to develop a method for the simultaneous quantification of methotrexate (MTX) samples extracted from human plasma and cerebrospinal fluid (CSF), using two-dimensional liquid chromatography (2D-LC). Furthermore, we intended to verify whether intravenous mannitol could increase MTX concentration in the CSF of patients.

Methods: The mobile phase of PUMP1 consisted of 10.

Article Synopsis
  • Nine compounds were extracted from ethanolic extracts, including one new alkaloid and one new pentacyclic triterpenoid, plus seven known alkaloids.
  • The structures of the new compounds were determined using 1D and 2D NMR spectroscopy and high-resolution mass spectrometry.
  • The antiproliferative activity of the compounds was tested against two cancer cell lines (LN229, A549), but compounds did not show significant activity.

Article Synopsis
  • The study analyzed the diagnostic and prognostic significance of Kruppel-like factor 5 (KLF5) and Runt-related transcription factor 1 (RUNX1) in patients with sepsis-induced acute kidney injury (SI-AKI) by measuring their levels in blood samples from 120 patients divided into SI-AKI and non-AKI groups.
  • Results indicated that both KLF5 and RUNX1 levels were significantly elevated in SI-AKI patients, proving their effectiveness in distinguishing these patients from those without kidney injury.
  • Furthermore, high levels of KLF5 and RUNX1 were linked to poor 28-day survival outcomes in SI-AKI patients, establishing their potential as independent prognostic markers for survival.*

Background: Skeletal muscle relaxants (SMRs) are widely used in treating musculoskeletal conditions. All SMRs, with the exception of baclofen and tizanidine, are on the list of 2023 American Geriatrics Society Beers Criteria for potentially inappropriate medication use in older adults. In our geriatric practice, off-label use of tizanidine as preemptive analgesia drove us to find recent advances in its pharmacology and therapeutics.

Identifying novel phytochemical secondary metabolites following classical pharmacognostic investigations is tedious and often involves repetitive chromatographic efforts. During the past decade, Ultra-High Performance Liquid Chromatography-Quadrupole Time of Flight-Tandem Mass Spectrometry (UHPLC-QToF-MS/MS), in combination with molecular networking, has been successfully demonstrated for the rapid dereplication of novel natural products in complex mixtures. As a logical application of such innovative tools in botanical research, more than 40 unique 3-oxy-, 3, 6-dioxy-, and 3, 6, 27-trioxy-steroidal saponins were identified in aerial parts and rhizomes of botanically verified .

Pomegranate extracts standardized to punicalagins are a rich source of ellagitannins including ellagic acid (EA). Recent evidence suggests that gut microbiota-derived urolithin (Uro) metabolites of ellagitannins are pharmacologically active. Studies have evaluated the pharmacokinetics of EA, however, little is known about the disposition of urolithin metabolites (urolithin A (UA) and B (UB)).
