Cell signaling pathways are enriched for biological processes crucial for cellular communication, response to external stimuli, and metabolism. Here, a cell signaling-focused CRISPR screen identified cytochrome c oxidase subunit 4 isoform 1 (COX4I1) as a novel vulnerability in acute myeloid leukemia (AML). Depletion of COX4I1 hindered leukemia cell proliferation and impacted in vivo AML progression.

Protein disulfide isomerase (PDI) regulates multiple protein functions by catalyzing the oxidation, reduction, and isomerization of disulfide bonds. The enzyme is considered a potential target for treating thrombosis. We previously developed a potent PDI inhibitor, CPD, which contains the propiolamide as a warhead targeting cysteine residue in PDI.

Polyurethane (PU) foam is widely used in industrial and civil fields, but it is highly flammable. An eco-friendly flame-retardant coating has been fabricated from sodium alginate (SA) and mica powder, it has been applied to PU foam using a facile direct dip coating method, followed by crosslinking with Ca and modification with polydimethylsiloxane (PDMS), respectively. The original porous network structure is maintained in the coated PU (SMPU) foam with a porosity of 90.

Human p97/VCP is a vital AAA ATPase (ATPase associated with diverse cellular activity) that plays critical roles in protein homeostasis by regulating autophagy, endosomal trafficking, and the ubiquitin-proteasome system. Global proteomics analysis of p97/VCP inhibition with CB-5083 has been performed in HCT116 colon cells. Here, we examined the impact of CB-5083 treatment in another cancer model, the HL-60 acute myeloid leukemia cell line, employing subcellular fractionation combined with label-free proteomics to analyze changes in protein levels across cytoplasmic, nuclear, and insoluble membrane protein compartments.

Background: Annual influenza epidemics lead to substantial public health burden, and pregnant people are vulnerable to severe outcomes. Influenza during pregnancy is hypothesized to increase risk of adverse birth outcomes, but population-based epidemiologic evidence remains limited and inconsistent.

Methods: We conducted a time-series analysis to estimate short-term associations between community-level seasonal influenza activity and daily counts of preterm births in Atlanta, United States from October 17th, 2010 to July 10th, 2017.

As obligate intracellular pathogens, viruses activate host metabolic enzymes to supply intermediates that support progeny production. Nicotinamide phosphoribosyltransferase (NAMPT), the rate-limiting enzyme of salvage nicotinamide adenine dinucleotide (NAD) synthesis, is an interferon-inducible protein that inhibits the replication of several RNA and DNA viruses through unknown mechanisms. Here, we show that NAMPT restricts herpes simplex virus type 1 (HSV-1) replication by impeding the virion incorporation of viral proteins owing to its phosphoribosyl-hydrolase (phosphoribosylase) activity, which is independent of the role of NAMPT in NAD synthesis.

To date, the development of high-performance n-type organic semiconductors has remained challenging due to the scarcity of highly electron-deficient π-conjugated structural units and the difficulty of controlling intermolecular packing in the thin-film state. In addition, there have been few reports on the use of dimer design to tune the optoelectronic properties of materials. Herein, we report new cyano-substituted fluoranthene imide-based dimers (F16 and F17) for small-molecule n-type organic semiconductors.

During the process of photoaging in the skin, Succinylated type I collagen has a significant effect on reversing the damage caused by UVB radiation, with the regulation of cellular ferroptosis being one of its important pathophysiological mechanisms. Specifically, Succinylated type I collagen reduces the expression of key cell cycle regulators P16, P21, and P53, as well as the ferroptosis-related factor Acyl-CoA Synthetase Long Chain Family Member 4 (ACSL4), induced by UVB radiation in cells and tissues. Meanwhile, it increases the expression of key factors Glutathione Peroxidase 4 (GPX4) and Solute Carrier Family 7 Member 11 (SLC7A11), which inhibit ferroptosis.

Article Synopsis
  • Multiple myeloma (MM) is a difficult-to-treat cancer characterized by chromosome instability and a reliance on the bone marrow environment, with p53 mutations associated with negative outcomes, yet some aggressive cases lack these mutations.
  • * Research shows that DNp73, an inhibitor of tumor suppressors, enhances drug resistance and cell proliferation in MM, with mechanisms still not fully understood.
  • * The study found that activation of NF-κB-p65 increases DNp73 levels, leading to more aggressive cancer behavior, drug resistance, and immune evasion through upregulation of MYCN and other immune-related genes.

  • - This study investigates the frequency and characteristics of MYD88 and CXCR4 mutations in patients with Waldenström macroglobulinemia (WM), aiming to find the best methods for clinical testing and their prognostic value across different treatments.
  • - A total of 385 symptomatic WM patients were tested using various genetic sequencing methods, revealing that MYD88 mutations occurred in 87.8% of patients, while CXCR4 mutations were found in 30.9%.
  • - Results indicated that while MYD88 and CXCR4 mutations are not significant prognostic factors in some treatment groups, they are critical for predicting outcomes in patients receiving Bruton's tyrosine kinase inhibitors (BTKis), surpassing the importance

  • The paper presents a low-power waveform designed to decrease the energy used during image refresh operations in Electrophoretic Displays (EPDs), aiming to lower the peak power of the system.
  • By activating particles before erasing them, the new method minimizes voltage polarity changes, which reduces overall power consumption.
  • Experimental results show that this approach decreases power fluctuation by 1.33% and energy fluctuation by 37.24%, improving display stability and reducing issues like screen flicker and ghosting.

Article Synopsis
  • Adeno-associated viruses (AAVs) are important tools used for delivering genes in research and clinical settings, but understanding how these viruses interact with cells can be challenging, especially for those that have been modified through directed evolution.
  • This study uses a human cell microarray platform to uncover how both natural and engineered AAVs connect with human cell receptors, discovering that AAV9 specifically interacts with interleukin 3 (IL3) and that engineered AAVs can interact with the low-density lipoprotein receptor-related protein 6 (LRP6), which might enhance their ability to cross the blood-brain barrier.
  • The findings also highlight potential side effects from engineered AAVs due to off-target tissue binding, paving the

Mammalian membrane proteins perform essential physiologic functions that rely on their accurate insertion and folding at the endoplasmic reticulum (ER). Using forward and arrayed genetic screens, we systematically studied the biogenesis of a panel of membrane proteins, including several G-protein-coupled receptors (GPCRs). We observed a central role for the insertase, the ER membrane protein complex (EMC), and developed a dual-guide approach to identify genetic modifiers of the EMC.
