SIRT3, a class III histone deacetylase, has been implicated in various cancers as a novel therapeutic target. In hepatocellular carcinoma (HCC), we previously reported that SIRT3 induced cell apoptosis by regulating GSK-3β/Bax signaling pathway. Downregulation of SIRT3 in HCC cells facilitates tumor cell survival.

Background/aim: The hepatitis B virus (HBV) infection is accompanied by the induction of oxidative stress, especially mediated by HBV X protein (HBx). Oxidative stress has been implicated in a series of pathological states, such as DNA damage, cell survival and apoptosis. However, the host factor by which cells protect themselves under this oxidative stress is poorly understood.

We previously reported that SIRT1, an NAD(+)-dependent deacetylase belonging to the class III histone deacetylases, enhances hepatitis virus B (HBV) replication by targeting the transcription factor AP-1. However, the potential antiviral effects of nicotinamide, a SIRT1 inhibitor, have not yet been explored. In this study, we show that nicotinamide exhibits potent anti-HBV activity with little cytotoxicity.

Background: The inhibitory Fc receptor, FcγRIIB, has emerged as a key negative regulator of B cell activation and as such is predicted to play an essential role in controlling antibody-mediated autoimmune diseases in humans. Recent studies have shown that crosslinking the FcγRIIB independently of the B-cell receptor (BCR) results in apoptosis in both mouse and chicken B cells. However, the human B cell subpopulations that are susceptible to BCR-independent, FcγRIIB-mediated regulation are not known.

Article Synopsis
  • Chronic liver diseases are common in China, with a study in Jilin revealing a 17.53% prevalence of elevated alanine aminotransferase (ALT) levels, primarily due to non-alcoholic fatty liver disease (NAFLD) and metabolic syndrome.
  • The study included 4,072 adults aged 18-79, showing that higher income, age, and certain health metrics like body mass index and fasting plasma glucose are significant risk factors for ALT elevation, particularly among female peasants and small businessmen.
  • Unlike metabolic disorders, elevated ALT levels related to hepatitis B or C infections were linked to gender, occupation, and age, highlighting the need for targeted health interventions in these demographics.

Article Synopsis
  • The study investigates the immune response in cirrhosis caused by hepatitis B virus (HBV) and hepatitis C virus (HCV), focusing on lymphocyte subsets and cytokine levels.
  • It finds that cirrhotic patients exhibit a lower proportion of specific immune cells (like CD8+ T cells and NK cells) and an increase in others (like CD4+ T cells and Treg cells) compared to healthy individuals.
  • Additionally, while Th2 cytokine IL-6 levels are elevated in both HBV and HCV cirrhosis, only HBV patients show an increase in the Th1 cytokine IFN-gamma, indicating a shared immune deficiency regardless of the virus type.
