Pirfenidone (PFD) is one of the first-line drugs for treating idiopathic pulmonary fibrosis, while directly delivering PFD to lung showed better efficiency. However, PFD is a non-glass former and easily precipitates into larger-sized crystals that are undesirable for pulmonary delivery. Hence, the fabrication of PFD particles with pulmonary delivery efficiency remains challenging.

  • Insomnia is a significant health issue, and delivering melatonin (MLT) nasally can improve its effectiveness by targeting the brain more directly.
  • The study created MLT embedded microparticles using different drying methods and evaluated their structure and release performance.
  • Results showed that microparticles made by spray drying were more effective for rapid release and better deposition in the olfactory region compared to those made by spray freeze drying.

Background: Neuroendocrine tumors (NETs) arise from the body's diffuse endocrine system. Coexisting primary adenocarcinoma of the colon and NETs of the duodenum (D-NETs) is a rare occurrence in clinical practice. The classification and treatment criteria for D-NETs combined with a second primary cancer have not yet been determined.

Active pharmaceutical ingredient (API) embedded dry powder for inhalation (AeDPI) shows higher drug loading and delivery dose for directly treating various lung infections. Inspired by the dandelion, we propose a novel kind of AeDPI microparticle structure fabricated by spray freeze drying technology, which would potentially enhance the alveoli deposition efficiency. When inhaling, such microparticles are expected to be easily broken-up into fragments containing API that acts as 'seed' and could be delivered to alveoli aided by the low density 'pappus' composed of excipient.

  • Fuqi Guben Gao (FQGBG) is a botanical drug used to treat nocturia linked to kidney-yang deficiency by combining several traditional medicinal plants, but its safety and pharmacological effects need further study.
  • Recent research on FQGBG demonstrated its effectiveness in reducing urination and improving immune organ damage in rat models of kidney-yang deficiency, while also assessing its safety in a clinical trial with healthy participants.
  • The results indicated significant improvements in symptoms with no major adverse events occurring during dose escalation, although some minor side effects were reported; pharmacokinetic analysis showed varying levels of certain compounds within FQGBG.

Article Synopsis
  • Endoscopy is crucial for detecting esophageal squamous cell carcinoma and precancerous lesions, but improving detection rates is still a challenge.
  • A new system using deep convolutional neural networks (CNNs) was developed and tested to enhance the detection of high-risk esophageal lesions (HrELs) during endoscopy, and showed significant improvement in detection rates.
  • In a clinical trial with over 3,100 patients, the CNN-assisted endoscopy group had a HrEL detection rate of 1.8%, compared to 0.9% in the control group, demonstrating greater sensitivity, specificity, and accuracy without any adverse events.

The immunosuppressive microenvironment caused by several intrinsic and extrinsic mechanism has brought great challenges to the immunotherapy of pancreatic cancer. We identified GFPT2, the key enzyme in hexosamine biosynthesis pathway (HBP), as an immune-related prognostic gene in pancreatic cancer using transcriptome sequencing and further confirmed that GFPT2 promoted macrophage M2 polarization and malignant phenotype of pancreatic cancer. HBP is a glucose metabolism pathway leading to the generation of uridine diphosphate N-acetylglucosamine (UDP-GlcNAc), which is further utilized for protein O-GlcNAcylation.

Objective: This study was designed to observe the effect of toll-like receptor 4 (TLR4)/nuclear factor kappa-B (NF-κB) pathway activity on sepsis-associated acute kidney injury (SA-AKI), thereby providing new considerations for the prevention and treatment of SA-AKI.

Methods: The rats were divided into Sham, cecal ligation and puncture (CLP), CLP + vehicle, and CLP + TAK-242 groups. Except the Sham group, a model of CLP-induced sepsis was established in other groups.

Deep cracking of bulky hydrocarbons on zeolite-containing catalysts into light products with high activity, desired selectivity, and long-term stability is demanded but challenging. Herein, the efficient deep cracking of 1,3,5-triisopropylbenzene (TIPB) on intimate ZSM-5@AlSBA-15 composites via tandem catalysis is demonstrated. The rapid aerosol-confined assembly enables the synthesis of the composites composed of a continuous AlSBA-15 matrix decorated with isolated ZSM-5 nanoparticles.

Primary biliary cholangitis (PBC) is a rare autoimmune liver disease with few effective treatments and a poor prognosis, and its incidence is on the rise. There is an urgent need for more targeted treatment strategies to accurately identify high-risk patients. The use of stochastic survival forest models in machine learning is an innovative approach to constructing a prognostic model for PBC that can improve the prognosis by identifying high-risk patients for targeted treatment.

To investigate the surgical efficacy of neurosurgery robot deep brain stimulation(DBS) in the treatment of elderly Parkinson's disease(PD). The clinical data of elderly patients (≥75 years) with PD who underwent neurosurgical robot-assisted DBS surgery in the Department of Neurosurgery of the General Hospital of Northern Theater Command from September 2016 to September 2022 were collected retrospectively. Operation time, electrode implantation duration, postoperative pneumocephalus volume, electrode implantation accuracy, the Tao's DBS surgery scale, perioperative complications were analyzed.

Pancreatic ductal adenocarcinoma (PDAC) is extremely malignant with limited treatment options. Deubiquitinases (DUBs), which cleave ubiquitin on substrates, can regulate tumor progression and are appealing therapeutic targets, but there are few related studies in PDAC. In our study, we screened the expression levels and prognostic value of USP family members based on published databases and selected USP10 as the potential interventional target in PDAC.

To investigate the effects of moderate intensity continuous training (MICT) and high intensity intermittent training (HIIT) on the ultrastructure of myocardium and soleus in rats with high fat diet, and to explore the mechanisms. 5-week-old male SD rats were randomly divided into normal diet quiet group (C), high-fat diet quiet group (F), high-fat MICT group (M) and high-fat HIIT group (H), with 8 rats in each group, and the fat content of the high-fat dietary feed was 45%. The M and H groups were given 12 weeks of treadmill running with an incline of 25°.

Intrinsic drug resistance mechanisms of tumor cells often reduce intracellular drug concentration to suboptimal levels. Epithelial-to-mesenchymal transition (EMT) is a pivotal process in tumor progression and metastasis that confers an aggressive phenotype as well as resistance to chemotherapeutics. Therefore, it is imperative to develop novel strategies and identify new targets to improve the overall efficacy of cancer treatment.
