Article Synopsis
  • Dual immune checkpoint blockade (ICB) using CTLA4 and PD-(L)1 inhibitors shows improved anti-tumor effectiveness and immune toxicity compared to PD-(L)1 inhibitors alone in advanced non-small-cell lung cancer (NSCLC) patients.
  • Patients with mutations in STK11 and/or KEAP1 genes benefit more from the combination treatment compared to those receiving only PD-(L)1 inhibitors, as shown in the POSEIDON trial.
  • The loss of KEAP1 serves as a strong predictor for the success of dual ICB, as it leads to a more favorable outcome by changing the tumor's immune environment to better engage CD4 and CD8 T cells for anti-tumor activity. *

Purpose: Various laboratory sources have recently achieved progress in implementing deep learning models on biomedical optical imaging of soft biological tissues. The highly scattered nature of tissues at specific optical wavelengths results in poor spatial resolution. This opens up opportunities for diffuse optical imaging to improve the spatial resolution of obtained optical properties suffering from artifacts.

Introduction: Although the 9-minute mean withdrawal time (m-WT) is often reported to be associated with the optimal adenoma detection rate (ADR), no randomized trials of screening colonoscopy have confirmed the impact of a 9-minute m-WT on adenoma miss rate (AMR) and ADR.

Methods: A multicenter tandem trial was conducted in 11 centers. Seven hundred thirty-three asymptomatic participants were randomized to receive segmental tandem screening colonoscopy with a 9-minute withdrawal, followed by a 6-minute withdrawal (9-minute-first group, 9MF, n = 366) or vice versa (6-minute-first group, 6MF, n = 367).

Article Synopsis
  • The study identifies a new variant in the WTIP gene associated with hypertrophic cardiomyopathy (HCM), suggesting its role in cardiac hypertrophy mechanisms.
  • Utilizing exome sequencing and other analyses, researchers found that knocking down WTIP in rat heart cells led to increased hypertrophy and altered calcium dynamics, which also appeared in patient-derived cells with the same genetic variant.
  • These findings highlight WTIP's critical involvement in cardiac health, showing that disruptions in its function can lead to significant heart conditions, particularly regarding calcium balance in heart cells.

Background: The availability of whole-genome sequencing data in large studies has enabled the assessment of coding and noncoding variants across the allele frequency spectrum for their associations with blood pressure.

Methods: We conducted a multiancestry whole-genome sequencing analysis of blood pressure among 51 456 Trans-Omics for Precision Medicine and Centers for Common Disease Genomics program participants (stage-1). Stage-2 analyses leveraged array data from UK Biobank (N=383 145), Million Veteran Program (N=318 891), and Reasons for Geographic and Racial Differences in Stroke (N=10 643) participants, along with whole-exome sequencing data from UK Biobank (N=199 631) participants.

Anosmia, the loss of smell, is a common and often the sole symptom of COVID-19. The onset of the sequence of pathobiological events leading to olfactory dysfunction remains obscure. Here, we have developed a postmortem bedside surgical procedure to harvest endoscopically samples of respiratory and olfactory mucosae and whole olfactory bulbs.

Rationale: Somatic overexpression in mice using an adeno-associated virus (AAV) as gene transfer vectors has become a valuable tool to analyze the roles of specific genes in cardiac diseases. The lack of atrial-specific AAV vector has been a major obstacle for studies into the pathogenesis of atrial diseases. Moreover, gene therapy studies for atrial fibrillation would benefit from atrial-specific vectors.

Rationale: Autosomal-dominant mutations in ryanodine receptor type 2 ( RYR2) are responsible for ≈60% of all catecholaminergic polymorphic ventricular tachycardia. Dysfunctional RyR2 subunits trigger inappropriate calcium leak from the tetrameric channel resulting in potentially lethal ventricular tachycardia. In vivo CRISPR/Cas9-mediated gene editing is a promising strategy that could be used to eliminate the disease-causing Ryr2 allele and hence rescue catecholaminergic polymorphic ventricular tachycardia.

The capacity to identify the unique functional architecture of an individual's brain is a crucial step toward personalized medicine and understanding the neural basis of variation in human cognition and behavior. Here we developed a cortical parcellation approach to accurately map functional organization at the individual level using resting-state functional magnetic resonance imaging (fMRI). A population-based functional atlas and a map of inter-individual variability were employed to guide the iterative search for functional networks in individual subjects.

A series of supramolecular assemblies of the type [Ru(L-L)2(L'-L)MX2)](n+) are reported where L-L is 2,2'-bipyridine (bipy), 4,4'-di-tetra-butyl-bipyridine (tbbipy) or 4,4'-diethoxycarbonyl-2,2'-bipyridine (dceb), L-L' is tetrapyrido[3,2-a:2',3'-c:3'',2''-h:2''',3'''-j]phenazine (tpphz), 2,2':5',2''-terpyridine (2,5-bpp), 2,2':6',2''-terpyridine, (2,6-bpp), 2,5-di(pyridine-2-yl)pyrazine (2,5-dpp) or 2,3-di(pyridine-2-yl)pyrazine (2,3-dpp), and MX2 is PdCl2, PtCl2 or PtI2. The photocatalytic behaviour with respect to hydrogen generation of these compounds and their ultrafast photophysical properties are discussed as a function of the nature of the peripheral ligands, the bridging ligands and the catalytic centre. The results obtained show how differences in the chemical composition of the photocatalysts can affect intramolecular photoinduced electron transfer processes and the overall photocatalytic efficiency.

Wnt signaling is one of the key oncogenic pathways in multiple cancers, and targeting this pathway is an attractive therapeutic approach. However, therapeutic success has been limited because of the lack of therapeutic agents for targets in the Wnt pathway and the lack of a defined patient population that would be sensitive to a Wnt inhibitor. We developed a screen for small molecules that block Wnt secretion.

This Letter presents a search for tb resonances in 1.04 fb(-1) of LHC proton-proton collision data collected by the ATLAS detector at a center-of-mass energy of 7 TeV. Events with a lepton, missing transverse momentum, and two jets are selected and the invariant mass of the corresponding final state is reconstructed.

This letter reports on a search for hypothetical heavy neutrinos, , and right-handed gauge bosons, , in events with high transverse momentum objects which include two reconstructed leptons and at least one hadronic jet. The results were obtained from data corresponding to an integrated luminosity of 2.1 fb collected in proton-proton collisions at [Formula: see text] with the ATLAS detector at the CERN Large Hadron Collider.
