Precise manipulation of genome structural variations holds great potential for plant trait improvement and biological research. Here we present a genome-editing approach, dual prime editing (DualPE), that efficiently facilitates precise deletion, replacement and inversion of large DNA fragments in plants. In our experiments, DualPE enabled the production of specific genomic deletions ranging from ~500 bp to 2 Mb in wheat protoplasts and plants.

Background: In the traditional computed tomography (CT) simulation process, patients need to undergo CT scans before and after injection of iodine-based contrast agent, resulting in a cumbersome workflow and additional imaging dose. Contrast-enhanced spectral CT can synthesize true contrast-enhanced (TCE) images and virtual noncontrast (VNC) images in a single scan without geometric misalignment. To improve work efficiency and reduce patients' imaging dose, we studied the feasibility of using VNC images for radiotherapy treatment planning, with true noncontrast (TNC) images as references and explored its dosimetric advantages compared to using TCE images.

Objectives: We evaluated spiritual coping levels among Chinese family caregivers of patients with advanced cancer and explored associated factors.
Immune checkpoint blockade is effective for some patients with cancer, but most are refractory to current immunotherapies and new approaches are needed to overcome resistance. The protein tyrosine phosphatases PTPN2 and PTPN1 are central regulators of inflammation, and their genetic deletion in either tumour cells or immune cells promotes anti-tumour immunity. However, phosphatases are challenging drug targets; in particular, the active site has been considered undruggable.

Gold nanoparticles (AuNPs) are cutting-edge platforms for combined diagnostic and therapeutic approaches due to their exquisite physicochemical and optical properties. Using the AuNPs physically produced by femtosecond pulsed laser ablation of bulk Au in deionized water, with a capping agent-free surface, the conjugation of functional ligands onto the AuNPs can be tunable between 0% and 100% coverage.
