Cholesterol homeostasis is pivotal for cellular function. Acyl-coenzyme A:cholesterol acyltransferase 1 (ACAT1), also abbreviated as SOAT1, is an enzyme responsible for catalyzing the storage of excess cholesterol to cholesteryl esters. ACAT1 is an emerging target to treat diverse diseases including atherosclerosis, cancer, and neurodegenerative diseases.

Article Synopsis
  • The phytopathogen studied has an ortholog of a surface layer protein (NpdA), but does not produce a traditional S-layer; the study aimed to confirm S-layer formation and its effects on virulence and other traits.
  • Experimental confirmations showed that the pathogen AAC00-1 does form an NpdA-based S-layer but did not exhibit any significant differences in virulence compared to the mutant strain in watermelon fruit tests.
  • NpdA orthologs were found in multiple strains of Betaproteobacteria and Gammaproteobacteria, suggesting that this new S-layer family plays a role in various Pseudomonadota, with potential implications for biofilm formation and material movement between cells.

Background: Doravirine is a non-nucleoside reverse transcriptase inhibitor recommended for the treatment of virologically suppressed and treatment naïve people living with HIV. The DRIVE-REAL study aimed to describe the characteristics, treatment patterns, and virological outcomes of doravirine users in a real-world cohort in the UK.

Methods: A retrospective, observational, multi-centre chart review was conducted for 300 adults living with HIV initiating doravirine-containing antiretroviral therapy.

Cholesterol is essential for cellular function and is stored as cholesteryl esters (CEs). CEs biosynthesis is catalyzed by the enzymes acyl-CoA:cholesterol acyltransferase 1 and 2 (ACAT1 and ACAT2), with ACAT1 being the primary isoenzyme in most cells in humans. In Alzheimer's Disease, CEs accumulate in vulnerable brain regions.
