The quantitative capabilities of a linear ion trap high-resolution mass spectrometer (LTQ-Orbitrap) were investigated using full scan mode bracketing the m/z range of the ions of interest and utilizing a mass resolution (mass/FWHM) of 15000. Extracted ion chromatograms using a mass window of +/-5-10 mmicro centering on the theoretical m/z of each analyte were generated and used for quantitation. The quantitative performance of the LTQ-Orbitrap was compared with that of a triple quadrupole (API 4000) operating using selected reaction monitoring (SRM) detection.

Quantitation of geometric isomers of a phosphodiesterase inhibitor was required to determine the extent of interconversion following dosing of a single isomer in preclinical pharmacokinetic studies. Assays were developed for the simultaneous determination of Compound A (Fig. 1), 6-[1-methyl-1-(methylsulfonyl)ethyl-8(3-{(E)-2-(3-methyl-1,2,4-oxadiazol-5-yl)-2-[4-(methylsulfonyl)phenyl]ethenyl}phenyl)quinoline] and its geometric Z-isomer, Compound B, in plasma using liquid chromatography-tandem mass spectrometry.

Enterohepatic recirculation (EHR) occurs via biliary excretion and intestinal reabsorption of a drug. Drug recycling through EHR can lead to a change in pharmacokinetic (PK) properties, such as reduced clearance (CL), extended half-life (T(1/2)) and increased plasma exposure (AUC). As a result, EHR may prolong the pharmacological effect of drugs.

Rifapentine is a cyclopentyl derivative of rifampin under development for the treatment of Mycobacterium tuberculosis and Mycobacterium avium complex infections. These studies were designed to investigate the disposition and biotransformation of single iv and oral doses of 14C-rifapentine in mice, bile duct-cannulated and uncannulated rats, and monkeys. Mass balance studies included 14C analyses of urine, feces, bile, cage wash, carcasses, and cage air collected for up to 120 hr postdose.
