Background: Exopolysaccharides (EPS) from probiotic bacteria like bifidobacteria, have gained considerable attention for the beneficial effects they exert in the gastrointestinal environment. Here, we investigated whether EPS isolated from Bifidobacterium longum subsp. infantis and Bifidobacterium adolescentis can interact with Toll-like receptors (TLRs) in a structure-dependent way and subsequently we investigated whether they influence cytokine-production in dendritic cells (DCs).

Introduction: Family planning and reproductive care are essential but complex aspects of lifecycle management for individuals with autosomal dominant polycystic kidney disease (ADPKD), given the potential genetic transmission and pregnancy-related complications. In this qualitative study, we studied the experiences and perspectives of patients with ADPKD and clinicians to identify areas for potential improvement in reproductive lifecycle care.

Methods: Focus group discussions (FGDs) were conducted in the Netherlands with patients with ADPKD, both men and women, who had children through varied reproductive choices; and clinicians, including (pediatric) nephrologists, obstetric gynecologists and geneticists.

Article Synopsis
  • Bifidobacteria produce complex exopolysaccharides (EPS) that have potential health benefits, but it's unclear how growth conditions, especially carbon sources, influence their structure.
  • Researchers studied the effects of two non-digestible carbohydrates, 2'-fucosyllactose (2'-FL) and galactooligosaccharides (GOS), on the growth of two Bifidobacteria species, B. adolescentis and B. infantis.
  • Results showed that B. adolescentis produced larger EPS molecules when exposed to GOS, while B. infantis thrived with 2'-FL and also created an additional polysaccharide with GOS, indicating the varied effects of different growth conditions on EPS production.

The intestinal epithelium is an important gatekeeper of the human body by forming a barrier for the luminal content of the intestine. The barrier function is regulated by a complex crosstalk between different cell types, including cells from the enteric nervous system (ENS). ENS is considered to influence gastrointestinal processes and functions, but its direct effect on epithelial barrier function remains to be confirmed.

Post-traumatic Parkinsonism (PTP) is a complex neurological disorder that is often associated with the occurrence of a traumatic brain injury (TBI). PTP can occur either in the acute or chronic phase of TBI. There is still uncertainty about the mechanisms provoking PTP, which can be the result of the acute blast itself or secondary neurodegenerative process occurring months to years post the acute trauma.

Base editing technologies enable programmable single-nucleotide changes in target DNA without double-stranded DNA breaks. Adenine base editors (ABEs) allow precise conversion of adenine (A) to guanine (G). However, limited availability of optimized deaminases as well as their variable efficiencies across different target sequences can limit the ability of ABEs to achieve effective adenine editing.

Pancreatic islet transplantation is proposed as a cure for type 1 diabetes mellitus (T1D). Despite its success in optimal regulation of glucose levels, limitations in longevity of islet grafts still require innovative solutions. Inflammatory stress post-transplantation and loss of extracellular matrix attribute to the limited β-cell survival.

The human diet requires a more plant-based approach due to the exhaustive effects animal-based foods have on the environment. However, plant-based proteins generally miss a few or have a lower variety in essential amino acids and are more difficult to digest. Subsequently they might be prone to fermentation by the microbiome in the proximal colon.

Unlabelled: During fertilization, mammalian sperm undergo a winnowing selection process that reduces the candidate pool of potential fertilizers from ∼10 -10 cells to 10 -10 cells (depending on the species). Classical sperm competition theory addresses the positive or 'stabilizing' selection acting on sperm phenotypes within populations of organisms but does not strictly address the developmental consequences of sperm traits among individual organisms that are under purifying selection during fertilization. It is the latter that is of utmost concern for improving assisted reproductive technologies (ART) because 'low fitness' sperm may be inadvertently used for fertilization during interventions that rely heavily on artificial sperm selection, such as intracytoplasmic sperm injection (ICSI).

  • Gut barrier disruption is a common issue in diseases like diabetes and IBD, leading to inflammation and changes in gut bacteria.
  • The study explores how secondary bile acids (BAs) and different types of dietary fibers, particularly pectin, affect gut epithelial cells' strength and tight junction gene expression.
  • Results show that certain unsulfated BAs and specific pectin types with different methylation and blockiness can help maintain gut barrier integrity, especially under high glucose conditions.

Article Synopsis
  • Researchers developed a co-culture model using an electric cell-substrate impedance sensing system (ECIS) to study how fermentation of non-digestible carbohydrates (NDCs) by gut bacteria affects gut barrier function.
  • They encapsulated intestinal bacteria in alginate to simulate a mucus layer and tested various levels of fermentable pectin, finding that increased pectin enhanced the protective effects on the barrier function of T84 gut epithelial cells.
  • The study demonstrated that higher pectin content led to improved gut health through increased production of beneficial short-chain fatty acids (SCFAs) and established a model for further research on bacteria and NDC interactions in gut barrier function.

Programmable DNA endonucleases derived from bacterial genetic defense systems, exemplified by CRISPR-Cas9, have made it significantly easier to perform genomic modifications in living cells. However, unprogrammed, off-target modifications can have serious consequences, as they often disrupt the function or regulation of non-targeted genes and compromise the safety of therapeutic gene editing applications. High-fidelity mutants of Cas9 have been established to enable more accurate gene editing, but these are typically less efficient.

Determining the optimal insemination moment for individual cows is complex, particularly when considering the effects of pregnancy on milk production. The effect of pregnancy on the absolute milk yield has already been reported in several studies. Currently, there is limited quantitative knowledge about the association between days post-conception (DPC) and lactation persistency, based on a lactation curve model, and, specifically, how persistency changes during pregnancy and relates to the days in milk at conception (DIMc).

Article Synopsis
  • Breastmilk is considered the best nutrition for infants because it contains unique nutrients that support gut, immune, and brain development; when breastfeeding isn't possible, infant formula is often used as an alternative.
  • Infant formulas typically lack human milk oligosaccharides (hMOs), which are important components found in breastmilk linked to various health benefits, including gut microbiome development and immune system support.
  • Recent studies indicate that certain hMOs, like 2'-fucosyllactose and lacto-N-neotetraose, positively impact infant development, but their use in formulas is limited by cost and complexity, highlighting the need for more research on effective hMO combinations.
