Comparison of tilt angles and provocative agents (edrophonium and isoproterenol) to improve head-upright tilt-table testing.

Patients with syncope underwent head-up tilt testing at 60 degrees and 80 degrees followed by edrophonium or isoproterenol challenge when indicated. The 80 degrees tilt protocol and edrophonium provocation were found to be as effective or more effective in eliciting neurally mediated syncope in susceptible patients.

Steel factor (c-kit ligand) stimulates the in vitro growth of immature CD3-/CD4-/CD8- thymocytes: synergy with IL-7.

The ability of Steel factor (SLF) to stimulate the growth of immature thymocytes alone and in combination with IL-7 was assessed. In suspension cultures of CD4-/CD8- thymocytes, SLF itself did not induce significant proliferation, but in combination with IL-7, it induced a greater response than did IL-7 alone. In fetal thymus lobe submersion cultures, SLF stimulated the growth of cells to a magnitude similar to that of IL-7 and the combination resulted in a synergistic response.

Chronic candidal meningitis: an uncommon manifestation of candidiasis.

Chronic meningitis is an uncommon manifestation of candidiasis. We present the case of an elderly woman who had symptoms such as headache, malaise, and fever for 8 months and was found to have Candida albicans meningitis, and we review 17 similar cases. An underlying illness or risk factor for candidiasis was present in only 13 (72%) of the 18 patients.

Inhibition of human red blood cell N-acetyltransferase.

Acetanilide and derivatives of benzoic acid, benzamide, salicylic acid, and salicylamide were tested as inhibitors of p-aminobenzoic acid N-acetyltransferase partially purified from peripheral blood of three rapid and two slow dapsone acetylator human volunteers. The 50% inhibition point of these compounds does not provide a kinetic basis for differentiating the rapid and slow dapsone acetylator phenotypes.

Beta 2-microglobulin-, CD8+ T-cell-deficient mice survive inoculation with high doses of vaccinia virus and exhibit altered IgG responses.

Transgenic mice lacking an intact beta 2-microglobulin (beta 2m) gene fail to express major histocompatibility complex (MHC) class I proteins on the cell surface and, as a result, are virtually devoid of CD4- CD8+ lymphocytes. These animals provide a unique model system for directly assessing the role of CD8+ lymphocytes in the modulation of viral infection in vivo. beta 2m- CD8- mice and their normal littermates were inoculated at the base of the tail with the WR strain of vaccinia virus and monitored for serum antibody and lesion formation.

Regulation of alloreactivity in vivo by IL-4 and the soluble IL-4 receptor.

Although numerous in vitro studies have demonstrated that cytokines are involved in the generation of alloreactive effector cells, the role of these regulatory substances in vivo is less well defined. We have recently cloned and expressed cDNAs encoding both membrane bound and soluble forms of the murine IL-4R. The effects of murine rIL-4 and a recombinant, soluble, extracellular portion of the murine IL-4R soluble(s) IL-4R on the generation of alloresponsiveness in vivo were evaluated by measuring the lymphoproliferative response to a localized injection of allogeneic cells and the survival of cardiac allografts.

Interleukin 2 stimulates serine phosphorylation of CD45 in CTLL-2.4 cells.

Ligation of interleukin 2 (IL2) is known to regulate both protein tyrosine and serine/threonine phosphorylation. A family of leukocyte transmembrane proteins whose cytoplasmic domain exhibits intrinsic protein tyrosine phosphatase activity is collectively called CD45 and is identified by a set of common cell surface epitopes. Although CD45 is known to be a phosphoprotein, it is not known how phosphorylation specifically regulates its function.

Prosthetic joint infections in the elderly.

Purpose: The aim of this study was to review risk factors, presenting symptoms and signs, organisms causing infection, treatment modalities, and outcomes in elderly patients with infection of prosthetic joints.

Patients And Methods: Patients over 60 years of age treated at the University of Michigan Hospital or Ann Arbor Veterans Administration Medical Center from 1977 to 1987 who had a diagnosis of prosthetic joint infection were reviewed. Early infections were defined as those occurring within 12 months of insertion of the prosthesis; late infections were those occurring more than 12 months after insertion.

Regulation of T cell proliferation by IL-7.

The regulation of murine T cell proliferation by IL-7 was investigated. Highly purified resting splenic T cells were induced to proliferate in a short term assay by IL-7 in the presence of the comitogen, Con A. The proliferation of these resting T cells showed both IL-2-dependent and -independent components as determined by the susceptibility of the response to the blocking effects of anti-IL-2 mAb.

Precursor frequency and lytic specificity of interleukin 2 and interleukin 4-responsive murine lymphocytes.

The response to IL-2 and IL-4 of several functionally defined lymphoid cell types was assessed at the individual responding cell level in limiting dilution experiments in which exogenous IL-2 or IL-4 was used to promote cellular activation and expansion. Splenic precursors that give rise to alloreactive CTL in LDA supplemented with IL-2 were approximately 3-fold more frequent than those detected in IL-4. Of special interest was the observation that cytolytic cultures arising in primary allogeneic LDA supplemented with IL-4 exhibited a greater degree of lytic specificity than those arising in IL-2.

Interleukin 7 stimulates growth of fetal thymic precursors of cytolytic cells: induction of effector function by interleukin 2 and inhibition by interleukin 4.

Interleukin 7 (IL-7) and interleukin 2 (IL-2) stimulate the outgrowth of distinct populations of thymocytes in lobe submersion cultures (LSC) established with day 12-14 murine fetal thymus. Analysis of the expression of cell surface markers in previous studies showed that IL-7 favors the expansion of a more immature population. In the experiments reported here, populations grown in IL-7 and IL-2 were found to differ functionally as assessed by the expression of cytolytic activity.
