Article Synopsis
  • Anatomical 3D-printed nasal casts are important for studying how drugs are delivered inside the nose and provide unique preclinical data not available from animal studies.
  • The study analyzed CT scans from 98 patients, revealing anatomical differences in the nasal cavity and identifying three distinct nasal geometry groups, leading to the creation of representative 3D models.
  • The research demonstrated that these anatomical variations significantly impact how well different medical devices deliver drugs, especially with nasal sprays and nebulizers, highlighting the need for diverse models to assess drug effectiveness in different populations.

The increasing prevalence of multidrug-resistant (PA) is a significant concern for chronic respiratory disease exacerbations. Host-directed drugs, such as flagellin, an agonist of toll-like receptor 5 (TLR5), have emerged as a promising solution. In this study, we evaluated the prophylactic intranasal administration of flagellin against a multidrug-resistant strain of PA (PA) in mice and assessed the possible synergy with the antibiotic gentamicin (GNT).

is a major hospital-associated pathogen that can cause severe infections, most notably in patients with cystic fibrosis (CF) or those hospitalized in intensive care units. Given its remarkable ability to resist antibiotics, eradication has grown more challenging. Therefore, there is an urgent need to discover and develop new strategies that can counteract -resistant strains.

This study describes the synthesis and evaluation of different imprinted hydrogels using ribavirin as template molecule. Ribavirin serves as a model molecule because it possesses a broad-spectrum antiviral effect against RNA viruses, which are expected as emerging viruses. The choice of monomers enables to stabilize the pre-polymerization complex and to synthesize biocompatible polymers.

Out of the 14 avian β-defensins identified in the genome, only 3 are present in the chicken egg, including the egg-specific avian β-defensin 11 (-AvBD11). Given its specific localization and its established antibacterial activity, -AvBD11 appears to play a protective role in embryonic development. -AvBD11 is an atypical double-sized defensin, predicted to possess 2 motifs related to β-defensins and 6 disulfide bridges.

is an opportunistic bacteria and a major cause of nosocomial pneumonia. has many virulence factors contributing to its ability to colonize the host. LoxA is a lipoxygenase enzyme secreted by that oxidizes polyunsaturated fatty acids.

Interleukin (IL)-22 plays a critical role in regulating the maintenance of the mucosal barrier. As airway epithelial regeneration is abnormal in cystic fibrosis (CF), we investigated IL-22 integrity in CF. We first demonstrated, using Il-22-/- mice, that IL-22 is important to prevent lung damage induced by the CF pathogen Pseudomonas aeruginosa.

The endotracheal tube (ETT) is an essential interface between the patient and ventilator in mechanically ventilated patients. However, a microbial biofilm is formed gradually on this tube and is associated with the development of ventilator-associated pneumonia. The bacteria present in the biofilm are more resistant to antibiotics, and current medical practices do not make it possible to eliminate.

Article Synopsis
  • - The enzyme cathepsin C (CatC) plays a role in activating inflammatory proteases in precursor immune cells, and inhibiting it early in bone marrow could help treat chronic inflammatory diseases.
  • - Research into CatC's maturation reveals it follows a multistep process managed by another enzyme, cathepsin S (CatS), and blocking CatS significantly reduces CatC activity while not entirely halting other neutrophil proteases.
  • - In patients with lung inflammation, activated neutrophils secrete large amounts of mature CatC, and ongoing clinical trials for CatS inhibitors aim to target CatC activity specifically, potentially reducing inflammation without affecting other important proteases.

K-ras mutations promote angiogenesis in lung cancer and contribute to the drug resistance of cancer cells. It is not clear whether K-ras mutated adenocarcinomas are sensitive to anti-angiogenic therapy with monoclonal antibodies (mAbs) that target vascular endothelial growth factor (VEGF). Anti-angiogenic mAbs are usually delivered systemically, but only a small proportion reaches the lung after intravenous injection.

Gallin is a 41-residue protein, first identified as a minor component of hen egg white and found to be antimicrobial against Escherichia coli. Gallin may participate in the protection of the embryo during its development in the egg. Its sequence is related to antimicrobial β-defensin peptides.

Rationale: Neutrophil serine proteases in cystic fibrosis (CF) lung secretions partially resist inhibition by natural and exogenous inhibitors, mostly because DNA impairs their control. Cationic polypeptides display the property of condensing DNA and retain antimicrobial properties. We hypothesized that DNA condensation by cationic polypeptides in CF sputum would result in a better control of CF inflammation and infection.

In dairy goats, treatments associating a progestogen and the equine chorionic gonadotropin (eCG) are the easiest way to induce and synchronize estrus and ovulation and to permit artificial insemination (AI) and/or out of season breeding. From the first treatment, the injection of eCG induces, in some females, the production of anti-eCG antibodies (Abs) that will interfere with the effectiveness of subsequent treatments. These anti-eCG Abs delay the preovulatory LH surge and the ovulation time, leading to poor fertility of the treated females.
