Article Synopsis
  • The development of a fluorescence lifetime-measuring flow cytometer allows for fast quantification of fluorescence lifetime in living or fixed cells, overcoming the limitations of traditional fluorescent imaging.
  • The instrument features a time-correlated single photon counting system integrated with microfluidics, achieving a maximum throughput of 3,000 particles per minute for statistical analysis on large cell populations.
  • This new technology enables sensitive detection of EGF receptor phosphorylation and explores dual-color fluorescence lifetime detection, paving the way for innovative applications in flow cytometry not previously possible due to earlier technological constraints.

We have successfully demonstrated video-rate CMOS single-photon avalanche diode (SPAD)-based cameras for fluorescence lifetime imaging microscopy (FLIM) by applying innovative FLIM algorithms. We also review and compare several time-domain techniques and solid-state FLIM systems, and adapt the proposed algorithms for massive CMOS SPAD-based arrays and hardware implementations. The theoretical error equations are derived and their performances are demonstrated on the data obtained from 0.
