RNA binding proteins (RBPs) containing intrinsically disordered regions (IDRs) are present in diverse molecular complexes where they function as dynamic regulators. Their characteristics promote liquid-liquid phase separation (LLPS) and the formation of membraneless organelles such as stress granules and nucleoli. IDR-RBPs are particularly relevant in the nervous system and their dysfunction is associated with neurodegenerative diseases and brain tumor development.

Article Synopsis
  • Most breast cancers are estrogen receptor (ERα) positive, and while endocrine therapy is commonly used, resistance often develops, making the enzyme fatty acid synthase (FASN) a target for treatment.
  • In studies with tamoxifen-resistant cell lines and patient samples, the FASN inhibitor TVB-3166 was shown to significantly decrease cell growth and tumor proliferation by reducing ERα levels and altering its location within the cell.
  • The findings indicate that targeting FASN with TVB-3166 could be an effective treatment strategy for patients with endocrine-resistant breast cancer, warranting further clinical exploration.

Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive Schwann cell-derived neoplasms that occur sporadically or in patients with neurofibromatosis type 1 (NF1). Preclinical research on sporadic MPNSTs has been limited as few cell lines exist. We generated and characterized a new sporadic MPNST cell line, 2XSB, which shares the molecular and genomic features of the parent tumor.

Inhibin is a heterodimeric TGFβ family ligand that is expressed in many cancers and is a selective biomarker for ovarian cancers; however, its tumor-specific functions remain unknown. Here, we demonstrate that the α subunit of inhibin (INHA), which is critical for the functionality of dimeric inhibin A/B, correlates with microvessel density in human ovarian tissues and is predictive of poor clinical outcomes in multiple cancers. We demonstrate that inhibin-regulated angiogenesis is necessary for metastasis.
