IL-6 signaling is involved in the pathogenesis of a number of serious diseases, including chronic inflammation and cancer. Targeting of IL-6 receptor (IL-6R) by small molecules is therefore an intensively studied strategy in cancer treatment. We describe the design, synthesis, and characteristics of two new bis-pentamethinium salts and (meta and para) bearing indole moieties.

Article Synopsis
  • - Neurodegenerative diseases like Alzheimer's and Parkinson's are increasingly challenging for healthcare systems, leading to a surge in research focused on their underlying causes.
  • - Studies link mitochondrial DNA problems (like oxidative damage and mutations) to the onset and progression of these diseases, indicating their significant role in neurodegeneration.
  • - The review also examines how factors such as neuroinflammation and protein misfolding contribute to these diseases, while highlighting the impact of different mitochondrial DNA haplogroups on disease risk and development.

The fluorescent probes based on Tröger's base motive with both coumarin and cyanine substitution 11-13 have been synthesized by multi-step synthesis in high overall yields. Intracellular localization of prepared probes have been tested using four different cell lines (HF-P4, BLM, U-2 OS and A-2058). Prepared probes have intensive green and red fluorescence.

Four novel fluorescent cores bearing a transformable functional group based on a π-expanded naphthalimide including a fused pyranone or furan ring have been prepared. Fluorescent probes LysoSers 13-16 for lysosomal targeting have been tested. Co-localization with a commercial lysosome specific marker confirmed that the LysoSers labeled the lysosomal compartment with high selectivity.
