A series of 8 europium (III) tris-beta-diketonates with common formula Eu(L)(3)Int, where L is acetyl acetone, thenoyltrifluoroacetone, benzoylacetone, dibenzoylmethane and Int is 1,10-phenanthroline or 2,2'-bipyridine, together with an analog without intercalating moiety (Eu(III)(acetyl acetone)(3)(H(2)O)(2)) were tested for cytotoxic activity in a panel of human tumor cell lines, using the MTT-dye reduction assay. The panel consisted of the leukemias HL-60, BV-173, SKW-3, K-562, LAMA-84 and the urinary bladder carcinoma 5637. The tested europium complexes with appended intercalator moieties exhibited profound cytotoxic effects with IC(50) values lower or comparable to those of the referent drug cis-DDP, whereby the 1,10-phenanthroline bearing compounds were invariably more active than the corresponding 2,2'-bipyridine analogs.

In vivo and in vitro experiments on the application of cell technologies to tissue defect closure were conducted; autologic mesenchymal stem cells on 3-demensional matrices were used. The authors analyze the results of the application of bioengineering tissue equivalents for the closure of soft tissue and upper airway defects after extensive resections performed in 52 oncological patients. Tissue equivalents with stem cells provide engraftment and long-term graft functioning; they also modify wound surface, thus stimulating wound epithelization.

Molecular virological studies of the field material collected in the epicenter of epizooty with high mortality among mute swans (Cygnus olor) in the area of the lower estuary of the Volga River (November 2005) could establish the etiological role of highly pathogenic influenza A (HPAI) virus of the subtype H5N1. Ten HPAI/H5N1 strains deposited at the State Collection of Viruses of the Russian Federation with the priority dated December 1, 2005 were isolated from the cloacal/tracheal swabs and viscera of sick and freshly died mute swans. Complete nucleotide sequences of all fragments of the genome of 6 strains have been deposited in the Gene Bank.

The excited-state dynamics of the DNA bisintercalator YOYO-1 and of two derivatives has been investigated using ultrafast fluorescence up-conversion and time-correlated single photon counting. The free dyes in water exist in two forms: nonaggregated dyes and intramolecular H-type aggregates, the latter form being only very weakly fluorescent because of excitonic interaction. The excited-state dynamics of the nonaggregated dyes is dominated by a nonradiative decay with a time constant of the order of 5 ps associated with large amplitude motion around the monomethine bridge of the cyanine chromophores.

Early defects due to the afflux of monocytic phagocytes into the inflammatory focus, rapid resorption of detritis and normalisation of correlation between contents of matrix proteinases and their inhibitors, stimulation of stromal cornea keratinocytes migration and their proliferation characterize repair of postburn defects in transplantation of the stromal equivalent. Repair against the background of stromal equivalent transplantation is of a histotypical character: collagen gel organizes and serves the basis for migration of the cornea fibroblasts and temporary matrix for growing new epithelium, while restored basal membrane is a derivative of cornea epithelium.

The interaction between the nicotinic acetylcholine receptor and Na+,K(+)-ATPase described previously was further studied in isolated rat diaphragm and in a membrane preparation of Torpedo californica electric organ. Three specific agonists of the nicotinic receptor: acetylcholine, nicotine and carbamylcholine (100 nmol/L each), all hyperpolarized the non-synaptic membranes of muscle fibers by up to 4 mV. Competitive antagonists of nicotinic acetylcholine receptor, d-tubocurarine (2 mcmol/L) or alpha-bungarotoxin (5 nmol/L) completely blocked the acetylcholine-induced hyperpolarization indicating that the effect requires binding of the agonists to their specific sites.

A quantum-dot-based saturable absorber has been demonstrated to initiate the generation of femtosecond pulses from a passively mode-locked solid-state laser. Control and tuning of the pulse duration from 58 ps to 158 fs was achieved. The 158 fs transform-limited pulses at 1280 nm are the shortest pulses that were produced from the Cr:forsterite laser passively mode locked by an InAs/InGaAs quantum-dot semiconductor saturable absorber mirror.

The regularities of 137Cs distribution and accumulation in organs and tissues of sheep on a farm located in the Chernobyl accidental zone were experimentally estimated. The distribution pattern of 137Cs concentration in organs and tissues is found to depend on the duration of the radionuclide uptake with the ration. During the first 24 h the highest 137Cs concentration is reported in the parenchymal organs; starting from day 30 muscular tissue and kidneys rank first, whereas in the other organs and tissues 137Cs concentration is 1.

Analysis of behavior of isolated epidermal keratinocytes demonstrated two dominant processes within the first two days of cultivation: formation of cell aggregates and their adhesion to the substrate. Keratinocyte spreading increased in the attached aggregates, where densely packed and well spread cells could be recognized in the aggregate center and periphery, respectively. Cell spreading and proliferation was observed in the following days.

Considerable hemorheological changes and depression of EEG parameters were revealed in rats with cerebral ischemia. Course peroral treatment with scarlet lightning extract in a daily dose of 150 mg/kg for 5 days reduced the severity of hemorheological disorders. It manifested in a decrease in whole blood viscosity, plasma viscosity, erythrocyte aggregation, and fibrinogen concentration and increase in deformability.

In experiments with isolated neuromuscular preparation of the rat diaphragm, selective blockade of alpha2 isoform of the Na,K-ATPase with ouabain (1 mcmol/L) induced steady depolarization of muscle fibers that reached a maximum of 4 mV, a decrease in amplitude of muscle fiber action potential, and prolonged raising and decline phases of the action potential. At the same time, the force, time to peak, and half relaxation time of the isometric muscle twitch were increased, as well as the area under the contraction curve. During continuous fatiguing stimulation (2/s), a more pronounced decline of contraction speed was observed in presence of ouabain; dynamics of the half-relaxation time remaining unchanged.

[Epidermal stem cells].

Izv Akad Nauk Ser Biol

July 2005

Epidermis contains a compartment of stem cells but currently there is no common ctiterion to recognize individual stem cells with any confidence. Epidermis appears to contain stem cells of different levels of maturity and it is very likely that the main repository of epidrmal stem cells is located in the hair follicle from which cells can emigrate into epidermis and also give rise to follicular and sebaceous keratinocytes. Epidermis consists of proliferative units containing stem and transit-amplifying cells, but the exact size of a proliferative unit cannot be measured accurately.

An improved epsilon expansion in the d -dimensional (d > 2) stochastic theory of turbulence is constructed at two-loop order, which incorporates the effect of pole singularities at d--> 2 in coefficients of the epsilon expansion of universal quantities. For a proper account of the effect of these singularities, two different approaches to the renormalization of the powerlike correlation function of the random force are analyzed near two dimensions. By direct calculation, it is shown that the approach based on the mere renormalization of the nonlocal correlation function leads to contradictions at two-loop order.

We compared some processes characteristic for both apoptosis and terminal differentiation of epidermal keratinocytes. It can be proposed that nonapoptotic programmed cell death takes place during differentiation of keratinocytes. Apoptosis and terminal differentiation of keratinocytes appear to be different processes but some similar molecular mechanisms are involved in these processes.
