Article Synopsis
  • Cyclin-F is a key protein involved in the regulation of the cell cycle and DNA damage response through its role in the SCF E3 ligase complex, which ubiquitinates various proteins.
  • Mutations in the gene CCNF, which encodes Cyclin-F, are linked to amyotrophic lateral sclerosis (ALS), but it's unclear how these mutations impact Cyclin-F's function.
  • Research identified HSP90 chaperone machinery as a new target of Cyclin-F, suggesting that impairment in Cyclin-F could disrupt chaperone functions and contribute to ALS pathogenesis.

Ubiquilin-2 (UBQLN2) is a ubiquitin-binding protein that shuttles ubiquitinated proteins to proteasomal and autophagic degradation. UBQLN2 mutations are genetically linked to the neurodegenerative disorders amyotrophic lateral sclerosis and frontotemporal dementia (ALS/FTD). However, it remains elusive how UBQLN2 mutations cause ALS/FTD.
