Article Synopsis
  • TNF superfamily member 15 (TL1A) acts as a ligand for TNFR superfamily member 25, which can enhance regulatory T cells (Tregs) when stimulated with an agonistic antibody (4C12).
  • A new soluble TL1A-Ig fusion protein was developed, showing the ability to rapidly proliferate both Foxp3(+) Tregs and conventional CD4(+) T cells while inhibiting the formation of new Tregs.
  • In vivo experiments demonstrated that TL1A-Ig significantly increased Treg populations, providing protection against allergic lung inflammation, indicating its potential as a therapeutic agent for controlling Treg proliferation.

Article Synopsis
  • TNFRSF25 (TNFR25) is a receptor found in CD4(+)FoxP3(+) Tregs, which have not been fully understood in terms of their function.
  • Using a specific antibody (4C12), researchers found that activating TNFR25 led to a significant increase in Treg levels within just four days, making them 30%-35% of all CD4(+) T cells in the blood of mice.
  • The expanded Tregs were highly effective in suppressing allergic lung inflammation in a mouse model for asthma, indicating TNFR25 plays a key role in Treg function and immune regulation.

Established tumors suppress antitumor immune responses and induce tolerance by incompletely characterized mechanisms, and this phenomenon is an important barrier to tumor immunotherapy. Single vaccination with tumor cells expressing gp96-Ig stimulates robust expansion of tumor-specific CTLs in tumor-naïve mice and this expansion is inhibited by established tumors. Interestingly, frequent vaccinations restore antitumor immune responses in the presence of established tumors.

The survival of viral mediated lymphomas depends upon constitutive nuclear factor kappa B (NF-kappaB) activity. AIDS-related human herpesvirus type 8-associated primary effusion lymphoma (PEL) responds poorly to chemotherapy and is almost invariably fatal. We have previously demonstrated that the antiviral combination of interferon alpha (IFN-alpha) and azidothymidine (AZT) induces apoptosis in PEL cell lines.
