The aim of this study was to develop a new series of isocryptolepines and evaluate their antiproliferative and antiestrogenic activities on cancer cells. A series of isocryptolepine derivatives were synthesized using developed one-pot photochemical, metal-free protocol, employing readily available 2-arylindoles as starting compounds. The resulting isocryptolepines demonstrated (sub)micromolar inhibitory activity against selected breast cancer cell lines.

Among these, there are two novel reactions (formal [1,3]-H migration and complete aromatization to an anthracene derivative) as well as photorearrangement and formal methane elimination. This diverse reactivity demonstrates the great potential of --diarylethenes in synthetic photochemistry.

The target compounds are furan analogues of phenanthrene formally accessed by isosteric replacement of the CHCH moiety with an oxygen atom. The straightforward and robust approach involving a [1,2]-aryl shift as a key step provides easy access to a wide range of naphtho[2,1-]furans with the possibility of late-stage functionalization.
