Host iron deficiency is protective against severe malaria as the human malaria parasite depends on bioavailable iron from its host to proliferate. The essential pathways of iron acquisition, storage, export, and detoxification in the parasite differ from those in humans, as orthologs of the mammalian transferrin receptor, ferritin, or ferroportin, and a functional heme oxygenase are absent in . Thus, the proteins involved in these processes may be excellent targets for therapeutic development, yet remain largely unknown.

The lysosomal degradation of macromolecules produces diverse small metabolites exported by specific transporters for reuse in biosynthetic pathways. Here we deorphanized the major facilitator superfamily domain containing 1 (MFSD1) protein, which forms a tight complex with the glycosylated lysosomal membrane protein (GLMP) in the lysosomal membrane. Untargeted metabolomics analysis of MFSD1-deficient mouse lysosomes revealed an increase in cationic dipeptides.

Article Synopsis
  • Proton-dependent oligopeptide transporters (POTs) play a key role in absorbing various dietary peptides and drugs, but understanding how they transport these different substrates has been a challenge.
  • Researchers presented 14 X-ray structures of the bacterial POT DtpB with diverse di- and tripeptides, revealing important information about the transporter's flexible binding site.
  • By studying binding affinities of over 80 peptides and using advanced techniques like molecular docking and machine learning, they found that peptides with compact hydrophobic residues are most effectively recognized by DtpB.

Cryogenic electron microscopy (cryo-EM) data represent density maps of macromolecular systems at atomic or near-atomic resolution. However, building and refining 3D atomic models by using data from cryo-EM maps is not straightforward and requires significant hands-on experience and manual intervention. We recently developed StarMap, an easy-to-use interface between the popular structural display program ChimeraX and Rosetta, a powerful molecular modeling engine.

A major complication of hemophilia A therapy is the development of alloantibodies (inhibitors) that neutralize intravenously administered coagulation factor VIII (FVIII). Immune tolerance induction therapy (ITI) by repetitive FVIII injection can eradicate inhibitors, and thereby reduce morbidity and treatment costs. However, ITI success is difficult to predict and the underlying immunological mechanisms are unknown.
