Mass cytometry permits the high dimensional analysis of cellular systems at single-cell resolution with high throughput in various areas of biomedical research. Here, we provide a state-of-the-art protocol for the analysis of human peripheral blood mononuclear cells (PBMC) by mass cytometry. We focus on the implementation of measures promoting the harmonization of large and complex studies to aid robustness and reproducibility of immune phenotyping data.

Test system was the rapid-growing solid Ehrlich carcinoma. Methylen-blue, hematoporphyrin derivative Halle and chlorophyllin produce a markedly reduction of the tumor weight.

Therefore, a new concept with utilisation of laser light is presented. After percutaneous renal punction the laser light fiber is placed antegrade into the ureter near the stenotic ostium and the stenosis is resected transurethrally under fluoroscopy onto the laser light spot.

The tumor-toxic effect of the photosensitizer HpD/Halle activated by laser radiation was demonstrated. Solid Ehrlich carcinoma was used as model. Investigations into the tumor growth, the HpD-distribution after different kinds of drug administration by means of fluorescence measurements, the phototoxicity and the reduction of tumor masses after HpD application and laser irradiation were carried out.
