Background: Deucravacitinib, an oral, selective, allosteric tyrosine kinase 2 (TYK2) inhibitor, is approved in multiple countries for treatment of adults with moderate to severe plaque psoriasis who are candidates for systemic therapy.

Objectives: To evaluate the safety and efficacy of deucravacitinib through 4 years in the Phase 3 POETYK PSO-1, PSO-2 and long-term extension (LTE) trials in psoriasis.

Methods: PSO-1 and PSO-2 (parent trials) randomized patients 1:2:1 to oral placebo, deucravacitinib 6 mg once daily (QD) or apremilast 30 mg twice daily.

The statistical problem of multiplicity is concerned with making protected multiple inferences and their valid interpretation in a particular study. Most discussions of multiplicity focus on the increase of type I error rate if testing is done without any adjustment, with only a few papers discussing its ramifications for type II errors/power. We provide a survey of main approaches to protected inference in biomedical studies, touching on procedures to control the family-wise error rate, false discovery rate, as well as false discovery exceedance probability.

Hormones are known to be involved in life-history trade-offs as systemic signals that establish functional links among traits and regulate key behavioural and physiological transitions between states in organisms. Although major functions of many steroid hormones such as testosterone are conserved among vertebrates, circulating concentrations vary widely both within and across species, and the degree to which observed hormone concentrations mediate life-history responses to environmental variation is less understood.
