Natural killer (NK) cells are present in the circulation and can also be found residing in tissues, and these populations exhibit distinct developmental requirements and are thought to differ in terms of ontogeny. Here, we investigate whether circulating conventional NK (cNK) cells can develop into long-lived tissue-resident NK (trNK) cells following acute infections. We found that viral and bacterial infections of the skin triggered the recruitment of cNK cells and their differentiation into Tcf1CD69 trNK cells that share transcriptional similarity with CD56TCF1 NK cells in human tissues.

In this analysis, the virtual specimen needs to be segmented to separate different parts or materials, a process that normally requires considerable human effort. In the Automated SEgmentation of Microtomography Imaging (ASEMI) project, we developed a tool to automatically segment these volumetric images, using manually segmented samples to tune and train a machine learning model.

A versatile processing software is required to handle the large amount of data concurrently collected in µ-IL and common MeV ion imaging measurements through the respective methodologies. The open-source freeware PyMca, with image processing and multivariate analysis capabilities, is employed to simultaneously process common MeV ion imaging and µ-IL data.
View Article and Find Full Text PDFA nonproprietary software package, "PyMca", primarily developed for X-ray fluorescence analysis offers an easy-to-use interface for calculating maps, by integrating intensity (of X-ray fluorescence, as well as any spectral data) over Regions Of Interest (ROI), by performing per pixel operations or by applying multivariate analysis. Here we show that, while initially developed to analyze hyperspectral two-dimensional (spatial) maps, this tool can be beneficial as well to anyone interested in measuring spectral variations over one or two dimensions, these dimensions being time, temperature, and so on. Different possibilities offered by the software (preprocessing, simultaneous analysis of replicas, of different conditions, ROI calculation, multivariate analysis, determination of reaction rate constant and of Arrhenius plot) are illustrated with two examples.
