The DNA intercalating agents 4'-(9-acridinyl-amino) methanesulfon-m-anisidide (m-AMSA) and adriamycin were studied by using filter elution methods to measure DNA single-strand breaks (SSB's), DNA-protein cross-links (DPC's), and double-stranded breaks (DSB's) in mouse leukemia L1210 cells. Both compounds produced SSB's and DPC's at nearly 1:1 ratios. The SSB's and DPC's were shown to be localized with respect to each other; this was inferred from the finding that filter assays based on protein adsorption completely prevented the elution of the DNA single-strand segments between SSB's.

2'-Deoxycoformycin (2'-dCF), a tight-binding inhibitor of adenosine deaminase, was administered to 26 pediatric patients with acute lymphoblastic leukemia in a Phase I study. Doses ranged from 0.25 to 1.

Surgical management of refractory ectopic supraventricular tachycardia arising in the left atrium requires cryoablation of the bundle of His and insertion of a permanent ventricular pacemaker. This study was designed to develop a technique to isolate the left atrium electrically from the remainder of the heart, so that an artificial pacemaking system would be unnecessary. Ten adult dogs were subjected to cardiopulmonary bypass and a standard left atriotomy was extended anteriorly across the mitral valve anulus between the right and left fibrous trigones.

To clarify the relationship between recipient presensitization and response to granulocyte (PMN) transfusion, we tested 187 non-HL-A matched donor-recipient pairs for the presence of antileukocyte antibody using granulocytotoxicity (G), lymphocytotoxicity (L), microleukoagglutination (M), and capillary leukoagglutination (C) assays. PMN increments per 10(11) transfused PMNs per square meter of body surface area, ascertained one hour following termination of transfusion, and the occurrence of nonhemolytic transfusion reactions, were correlated with the assay results. Although circulating anti-donor-leukocyte antibody was detected in 52 per cent of recipients, there was no statistically significant relationship between the presence of these antibodies and either PMN recovery or incidence of transfusion reaction.

A review of the histology, clinical findings and results of therapy in 9 females with endodermal sinus tumor (EST) is presented. Five patients had histologically pure EST; 4 had EST mixed with other germ cell components. The site of primary tumor was the ovary in 8 of the 9 females; the remaining patient with an extraovarian primary represents the first reported case of EST arising in the vulva.
