The ability of aquatic mesofauna representatives involved in trophic chains to sorb and accumulate toxicants is important for understanding the functioning of aquatic ecosystems and for fishing industry. This study investigated the capacity of marine amphipod and freshwater amphipods and to absorb the DNA-alkylating agent methyl methanesulfonate (MMS). The presence of alkylating agents in the environment and in the tissues of the amphipods was determined using whole-cell lux-biosensor MG1655 pAlkA-lux, in which the genes from , enabling the luminescence of the cell culture, are controlled by the P promoter of DNA glycosylase.

Among the most promising are derivatives of triphenylphosphonium, which can protect the infected organs of mammals and heal damaged cells as mitochondria-targeted antioxidants. In addition to the antioxidant action, triphenylphosphonium derivatives exhibit antibacterial activity.

In this work, the genotoxic properties of dcpVNB were studied using whole-cell -biosensors based on and . It was shown that the addition of dcpVNB to bacterial cells leads to the appearance of DNA damage inducing the SOS response and Dps expression with slight activation of the OxyR-mediated response to oxidative stress.

Hybrid plasmids pNK-DinC, pNK-AlkA, and pNK-MrgA have been constructed, in which the reporter genes are transcribed from the stress-inducible promoters of : the SOS promoter P, the methylation-specific response promoter P, and the oxidative stress promoter P. The luminescence of based biosensors specifically increases in response to the appearance in the environment of such common toxicants as mitomycin C, methyl methanesulfonate, and HO.
View Article and Find Full Text PDFLuxR and LuxR1 and LuxR2 regulatory proteins are quorum sensing transcriptional (QS) activators, inducing promoters of genes in the presence of an autoinducer (3-oxo-hexanoyl-l-homoserine lactone). In the cells, genes are regulated by HNS, CRP, LitR, etc. Here we investigated the role of the expression level in LuxI/R QS system functionality and improved the whole-cell biosensor for autoinducer detection.
