Introduction: Oxidative state, a risk factor for several diseases, is increased by habitual conventional cigarette (CC) smoking. Reports have demonstrated that heat-not-burn cigarettes (HNBCs), which have recently become popular among smokers, generate less oxidative state than CC in smokers with a long smoking history. However, no previous study has examined oxidative state in young HNBC users.

Article Synopsis
  • This study compares the effects of consuming soy protein isolate (SPI) versus soy peptide (PEP) before a glucose tolerance test on stomach emptying, insulin, and blood sugar levels.
  • Nine healthy young individuals participated in two experiments, consuming either SPI or PEP before the test to assess their glycemic response and gastric emptying.
  • Results showed that PEP led to higher insulin levels and faster gastric emptying than SPI, suggesting that PEP may better regulate blood sugar after meals.

Geranyl diphosphate (GPP) is the direct precursor of all monoterpenoids and is the prenyl source of many meroterpenoids, such as geranylated coumarins. GPP synthase (GPPS) localized in plastids is responsible for providing the substrate for monoterpene synthases and prenyltransferases for synthesis of aromatic substances that are also present in plastids, but GPPS activity in localizes to the cytosol, in which GPP is utilized for the biosynthesis of naphthoquinone pigments, which are shikonin derivatives. This study describes the identification of the cytosol-localized GPPS gene, , through EST- and homology-based approaches followed by functional analyses.

Most of the α-halo carbonyl (AHC) compounds tend to be predicted as mutagenic by structure-activity relationship based on structural category only, because they have an alkyl halide structure as a structural alert of mutagenicity. However, some AHC compounds are not mutagenic. We hypothesized that AHC reacts with DNA by S2 reaction, and the reactivity relates to mutagenicity.

The cause of pleural effusion remains uncertain in approximately 15% of patients despite exhaustive evaluation. As recently described immunoglobulin (Ig)G4-related disease is a fibroinflammatory disorder that can affect various organs, including the lungs, we investigate whether idiopathic pleural effusion includes IgG4-associated etiology. Between 2000 and 2012, we collected 830 pleural fluid samples and reviewed 35 patients with pleural effusions undiagnosed after pleural biopsy at Yamaguchi-Ube Medical Center.

Objective Acute exacerbations of idiopathic pulmonary fibrosis (AE-IPF) are fatal episodes of acute respiratory worsening of unknown etiology. Previous studies on acute respiratory distress syndrome have shown that direct hemoperfusion with a polymyxin B-immobilized fiber column (PMX-DHP) can have a beneficial effect on the respiratory status. This retrospective study investigated the prognosis and survival outcome of patients with AE-IPF who underwent PMX-DHP.

Glycosylation of the IgG-Fc is essential for optimal binding and activation of Fcγ receptors and the C1q component of complement. However, it has been reported that the effector functions are down-regulated when the Fc glycans terminate in sialic acid residues and that sialylated IgG mediates anti-inflammatory effects of intravenous immunoglobulin (IVIG). Although recombinant IgG is hypo-sialylated, Fc sialylation is shown to be markedly increased when a mouse/human chimeric IgG3 Phe243Ala (F243A) variant is expressed in Chinese hamster ovary (CHO)-K1 cells.

Background: To investigate more practical handling of Borg's ratings of perceived exertion (RPE) and category-ratio scale of RPE (CR-10), we evaluated interrelationships between RPE, CR-10, and blood lactate concentrations (bLa) during incremental treadmill running tests for young females with different aerobic fitness levels.

Methods: Oxygen consumption, heart rate, bLa, RPE, and CR-10 were measured from distance runners (DR; n = 15), race walkers (RW; n = 6), and untrained females (UT; n = 11). These variables corresponding to the lactate threshold (LT) and onset of blood lactate accumulation (OBLA) were compared among these groups.

Purpose: Whether bevacizumab enhances the effect of the epidermal growth factor receptor (EGFR) inhibitor gefitinib on EGFR mutant non-small cell lung cancers (NSCLCs) remains unknown. We conducted a phase II trial to investigate the efficacy and safety of gefitinib when combined with bevacizumab as first-line therapy in patients with advanced NSCLC harboring EGFR gene mutations.

Methods: In this trial, 42 patients with a performance status of 0 to 2 received gefitinib (250 mg/d) and bevacizumab (15 mg/kg, every 3 weeks).

Background: Although cisplatin-based chemotherapy combined with thoracic irradiation (TRT) is a standard treatment for unresectable, locally advanced non-small cell lung cancer (NSCLC), this treatment outcome has remained unsatisfactory. We had previously conducted a phase I trial of cisplatin plus S-1, an oral 5-fluorouracil derivative, and TRT, which were safe and effective.

Methods: In this phase II trial, 48 patients with stage III NSCLC received cisplatin (40mg/m(2) on days 1, 8, 29 and 36) and S-1 (80mg/m(2) on days 1-14 and 29-42) and TRT (60Gy).

Article Synopsis
  • Thoracic irradiation is commonly used for elderly patients with advanced non-small-cell lung cancer, but outcomes are often poor; a previous trial showed promise by combining it with S-1, an oral chemotherapy drug.
  • In a phase II trial involving 30 patients aged 76 and older, the combined treatment of S-1 and thoracic radiation resulted in a 63% partial response rate, with median progression-free and overall survival times of 13.0 and 27.9 months, respectively.
  • While the study did not meet its primary endpoint of response rate, the treatment was generally well-tolerated with mostly mild side effects, suggesting it could be a viable option for this patient group.

Acute exacerbation of idiopathic pulmonary fibrosis (AE-IPF) is characterized by severe worsening dyspnea of unknown etiology and high mortality without effective treatment. Recently, direct hemoperfusion with polymyxin B (PMX)-immobilized fiber cartridge (PMX-DHP) has been reported to improve pulmonary oxygenation and survival in patients with AE-IPF although its mechanism of action remains unknown. To gain insights into the pathobiology of AE-IPF through the beneficial effects of PMX-DHP, we analyzed the profile of cytokines adsorbed onto PMX-fibers used in 9 AE-IPF patients.

Hybrid liposomes (HLs), composed of l-α-dimyristoylphosphatidylcholine (DMPC) and polyoxyethylene(23) dodecyl ether, have selectively inhibited the growth of human hepatocellular carcinoma (HCC) cells without affecting normal hepatocytes to trigger apoptosis via caspase-3 activation. Furthermore, HLs distinguished between the HCC and normal cells which had higher and lower membrane fluidities respectively, then fused and accumulated preferentially into the membranes of HCC cells. It is noteworthy that the anti-cancer activity of HLs correlated well with the fluidity of cell membranes for HCC and other cancer cells.

Expression of the transcription factor FOXP3 characterizes regulatory T cells (Tregs) that engage in the maintenance of immunological self-tolerance and immune homeostasis. Intra-tumoral accumulation of Tregs is associated with unfavorable prognosis in several kinds of cancers. Recently, expression of FOXP3 and its association with prognosis have also been shown in some cancer cells in clinical studies.
