Relapse or progressive disease after chimeric antigen receptor T-cell (CAR-T) treatment remains a major issue for poor-risk aggressive large B-cell lymphoma. However, limited data are available on post-CAR-T use of polatuzumab vedotin. Here we describe the case of a patient with diffuse large B-cell lymphoma (DLBCL) who experienced relapse three months after CD19-directed CAR-T therapy with tisagenlecleucel.

CD147 is an immunoglobulin-like receptor that is highly expressed in various cancers and involved in the growth, metastasis, and activation of inflammatory pathways via interactions with various functional molecules, such as integrins, CD44, and monocarboxylate transporters. Through screening of CD147-targeting antibodies with antitumor efficacy, we discovered a novel rat monoclonal antibody 147D. This humanized IgG4-formatted antibody, h4147D, showed potent antitumor efficacy in xenograft mouse models harboring the human PDAC cell line MIA PaCa-2, HCC cell line Hep G2, and CML cell line KU812, which featured low sensitivity to the corresponding standard-of-care drugs (gemcitabine, sorafenib, and imatinib, respectively).

Mogamulizumab (MOG), a humanized monoclonal anti-CCR4 antibody, exerts strong antibody-dependent cellular cytotoxic effects on CCR4-positive adult T-cell leukemia/lymphoma (ATLL) cells. As CCR4 is highly expressed on regulatory T cells as well as ATLL cells, pre-transplant MOG induces severe graft-versus-host disease (GvHD). However, limited data are available on post-transplant use of MOG for relapsed ATLL.

Purpose: To reveal the effect of drug-eluting beads transarterial chemoembolization and Lipiodol transarterial chemoembolization on portal perfusion, and to identify factors predisposing portal vein damage after transarterial chemoembolization, based on evaluation by computed tomography during arterial portography.

Material And Methods: This retrospective cohort analysis included 49 patients with hepatocellular carcinoma who underwent transarterial chemoembolization and preprocedural/follow-up computed tomography during arterial portography between October 2013 and April 2015. The preprocedural and follow-up computed tomography during arterial portography were compared to identify the following new changes suggestive of portal vein damage in the follow-up computed tomography during arterial portography: small perfusion defects, large perfusion defects, and narrowing/disappearance or portal vein obstruction.

Objective: The Oncotype DX (ODX) estimates the 10-year risk of metastasis or recurrence of breast cancer and indicates whether chemotherapy is likely to be effective; however, the high cost of this test may limit its use for patients. The aim of this study was to evaluate the potential of preoperative imaging using mammography (MMG), ultrasonography (US), and dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI), and positron emission tomography/computed tomography (PET/CT) metabolic parameters in predicting the ODX recurrence score (ODXRS), which prognosticates estrogen receptor-positive (ER +)/human epidermal growth factor receptor 2-negative (HER2-) breast cancer.

Methods: This retrospective study was conducted on 51 patients with ER+/ HER2- early-stage breast cancer with preoperative images available.

Objectives: To compare the predictive ability of liver fibrosis (LF) by CT-volumetry (CTV) for liver and spleen and extracellular volume fraction (ECV) for liver in patients undergoing liver resection.

Methods: We retrospectively analysed 90 consecutive patients who underwent CTV and ECV. Manually placed region-of-interest ECV (manual-ECV), rigid-registration ECV (rigid-ECV), and nonrigid-registration ECV (nonrigid-ECV) were calculated as ECV(%) = (1-haematocrit) × (ΔHU/ΔHU), where ΔHU = subtraction of unenhanced phase from equilibrium phase (240 s).

Article Synopsis
  • A study analyzed lifespan changes in patients who died from hematological cancers in Japan from 1990 to 2015 using the average lifespan shortened (ALSS) measure.
  • Data was collected from the World Health Organization, calculating years of life lost (YLL) against expected lifespans based on Japanese life tables.
  • Results indicated a significant improvement in the lifespan of these patients, with notable decreases in years of life lost over the years, especially for women with leukemia and men with non-Hodgkin lymphoma.

Article Synopsis
  • A multicenter study was conducted to examine if a comprehensive geriatric assessment (CGA) can predict adverse events from chemotherapy in elderly patients with diffuse large B-cell lymphoma (DLBCL).
  • The study involved 86 patients aged 65 and older, who underwent a baseline CGA assessing daily activities, mood, nutrition, comorbidities, and cognitive function; results showed that impaired instrumental activities of daily living (IADL) were linked to more severe chemotherapy toxicity.
  • Findings indicated that patients classified as independent had a higher 4-year survival rate (72.7%) compared to dependent patients (56.9%), suggesting CGA may help predict serious chemotherapy-related complications in this age group.

Background: Reduction of blood group ABO antigens on red blood cells (RBCs) is well known in patients with leukemias, and this reduction of ABO expression is strongly associated with DNA methylation of the ABO promoter. Previously, we reported a two-nucleotide deletion in RUNX1 encoding an abnormally elongated protein lacking the trans-activation domain in a patient with myelodysplastic syndrome (MDS) showing A-antigen loss on RBCs. This prompted us to investigate the underlying mechanism responsible for A-antigen reduction on RBCs in another patient with MDS.

Endovascular treatment is prevalent as a primary treatment for coronary and peripheral arterial diseases. Although the introduction of drug-eluting stents (DES) dramatically reduced the risk of in-stent restenosis, stent thrombosis persists as an issue. Notwithstanding improvements in newer generation DES, they are yet to address the urgent clinical need to abolish the late stent complications that result from in-stent restenosis and are associated with late thrombus formation.

Article Synopsis
  • * Researchers explored the transcriptional regulation of the gene OBP2B, located near the ABO gene, specifically focusing on a regulatory site 22.6 kb downstream in gastric cancer cells.
  • * Using CRISPR/Cas9 to create deletions, they found that mutations led to reduced levels of both OBP2B and ABO transcripts, suggesting that OBP2B may be co-regulated by the ABO site's activity.

Transplant acquired food allergy (TAFA) is a well-known complication following pediatric liver transplantation, but post-cord blood transplantation (post-CBT) TAFA has rarely been reported. Here, we describe a case of new-onset food anaphylaxis after CBT in an adult patient that demonstrates that post-CBT allergen-challenge is not a risk for long-term allergic sensitization even in adult recipients. The patient was a 39-year-old Japanese man with aggressive NK cell leukemia.

Rod-like inclusions have rarely been described in cases of monoclonal gammopathy of renal significance or multiple myeloma. The key finding in our case is the strong κ-light chain positivity of rod-like inclusions in plasma cells, which confirms their immunoglobulin nature. Therapeutic benefit of bortezomib was also observed.
